| SIPI-A.3201is a strain,preserved in our laboratory,which may produce fusidic acid. In thispaper, some researches were carried out of which include strain improvement, optimization ofthe processes of fermentation, separation and purification.After fusidic acid was determined and verified by HPLC and LC-MS in the broth fromstrain SIPI-A.3201proserved in Lab, natural selection and UV mutagenesis were carried out.The high-yield mutant SIPI-A.3202, whose fermentation level of fusidic acid was up to180μg/mL.The relationship of the productivity and morphology between high-and lower-strains werestudied. The results showed that the amount and speed of spores fomed during fermentation wereinversely proportional to the biosynthesis of fusidic acid.Fermentation medium and cultivation condition were optimized. The productivity of mutantSIPI-A.3202under the optimized medium and cultivation condition increased about three timescompared to the parent strain under original medium and condition.Fusidic acid was separated and purified from the broth according to the following process.The pH of broth was adjusted to2~3by adding oxalic acid, and then the cells were harvested bycentrifugation and extracted by95%ethanol. The extracted phase was concentrated andseparated and purified by macroporous adsorption HZ-818. The fusidic acid with purity of90%above was obtained. |
