| In our country, there are only Vancomycin and Teicoplanin which are used for clinicaltreatment of Methicillin-resistant Staphylococcus aureus infection. It will result in thebacterial drug resistance if the two medicines are long-term or frequently used for thetreatment. Fusidic Acid (Fusidic Acid, woven chains spore acid antibiotics) can be used aseffective supplement and substitute of Vancomycin and Teicoplanin for its wide range ofapplication, low toxicity, and excellent anti-drug resistance.In this topic, we focus on Strains screening of original strains and optimization offermentation process, so that we can get better titer to help the Enterprise reduce the costand increase economic benefit. By observation and monitoring for Fusidic Acid strains, wefound the growth forms and condition of the bacteria at different growth stage, and thebacterial growth curve. Then we did some experiment based on uniform design of themedium with different composition such as carbon, nitrogen, trace elements, and we gotthe final result as following:After several experiment and data analysis, we got the best formulation of shake flaskexperiments: sugar4.3%, dextrin2%, yeast1%, peptone3%, biological nitrogen0.59%,and magnesium sulfate0.2%and potassium dihydrogen phosphate0.15%, and the averageefficacy detected is907.62μg/ml, which is334%improved than the original titer of271.8μg/ml.The optimization of the fermentation formula are: sugar4.3%, dextrin2%, yeast1%,peptone3%, biological nitrogen0.59%, and magnesium sulfate0.2%and potassiumdihydrogen phosphate0.15%.The optimal the fermentation condition are: temperature:25°C; ventilation:1:1V/V/Min; inoculums size:5%; speed up the stir from500rpm to600rpm after the delayphase; adjust the pH by ammonia to keep it6.00.Determine the fermentation process are: carbohydrate supplementation at96h;supplementation amount is1%; supplementation rate is60ml/L.The final average titer detected is1105.6μg/ml, which is4times of the original271.8μg/ml. Through the experiment in this text, we can verify the practicability of the new strains production. We hope to go further to promoting the use of it, so that can helpenterprises expand production and increase econo mic efficiency. |
