| Colla Corii Asini, also known as the donkey-hide gelation, which was recor-ded in “Shen Nong’s Materia Medicaâ€. E-jiao is made from the skin of Equusasinus which is stewed, and it has been used for more than2,500years. It contai-ns collagen, amino acids, peptides, dermatan sulfate, selenium, zinc and othertrace elements, which is thought sweet, smooth in the chinese medicine. Notonly can E-jiao be used for treating blood deficiency colorosis, hematemesis, he-maturia and other aspects, but also applied to the foodstuff fields.The traditional Colla Corii Asini is neat rectangular, wide brown, andbrittle block. It needs to be smashed and stewed with water before drinking. TheE-jiao powder and E-jiao granules are producted in recent years which isconvenient to drink and carry, but when drinking we need to add rock sugar toconceal the special smell. In order to prepare the stable quality, efficacy andreliable, quickly crumbling of Colla Corii Asini effervescent tablets, thedissertation studied the formula of Colla Corii Asini effervescent granules, theoptimum temperature and time for drying, the best preparation formula andmethod of Colla Corii Asini effervescent tablets. The results were as follows.The preparation of Colla Corii Asini effervescent granules was studiedfirstly. The optimal thinner for adding was lactose, the organic acid was citricacid, the adhesive was6%PVP alcohol solution, the sweetener was proteinsugar. There were two formulas for preparation of E-jiao effervescent granules.The first formula was Colla Corii Asini70.7%, lactose23.6%, citric acid2.8%,sodium bicarbonate2.5%, protein sugar0.4%, and some6%PVP alcoholsolution. The second formula was Colla Corii Asini60.8%, lactose20.3%, citricacid9.7%, sodium bicarbonate8.8%, protein sugar0.4%, and some6%PVPalcohol solution. In order to study the effect of different temperature and time for drying onwater content of Colla Corii Asini effervescent granules, the50-60celsiusdegree and3-5hours were selected. The result showed that the optimum dryingcontidion was55celsius degree for roasting four hours or60celsius degree forroasting three hours.The production craft of E-jiao effervescent tablets was studied. The resultshowed that the optimum lubricant for adding was PEG6000, and the optimalformula was Colla Corii Asini60.8%, lactose20.3%, citric acid9.7%, sodiumbicarbonate8.8%, protein sugar0.4%, some6%PVP alcohol solution, andPEG60003%. Finally, by comparing the five preparation technology methodsfor E-jiao effervescent tablets which were acid-base respectively tablettingmethod, acid-base mixed tabletting method, powder compression method, semidry granules tabletting method, and dry granules tabletting method, theprefertable method was acid-base respectively tabletting method.The quality standard of E-jiao effervescent tablets was studied. The resultsshowed that the hardness, friability, pH, foaming, disintegration time, watercontent, weight difference, protein content, microbial limits, and harmful metalsindices all were correspond with the regulations of pharmacopoeia.The pharmaodynamics of E-jiao effervescent tablets was observed byhemopoiesis function, immune function, anti-fatigue and anti-hypoxia. Theresult indicated that E-jiao effervescent tablets had the curative effect ofphenyhydrazine hydrochloride and cyclophosphamide anemia mice, and it couldenhance the ability of anti-fatigue and anti-hypoxia of mice. In addition, it alsocould improve the cellular immunity and non-specific immunity in mice. |
