Food safety concerns everyone’s health and has attracted more attention around the world. With global trade integration, the theme of the development of food tends to be more healthysafety, sanitary and nutrition.But it makes a great deal of material damage because of microbial activities, insect pest and the problems existing in processing and storage of products. Irradiation technology as one of physical means can kill the pathogenic microorganisms, saprogenic bacteria and parasites thoroughly in meat. Then it can improve the hygiene quality, enhance the safety of meat and protect people’s health.But food irradiation technology is developing rapidly with imbalance in recent years.Meanwhile countries all the world put their own interests on top priority and fix rules benefit for themselves. But it has not established the detection method for irradiated food containing lipid in our country. And the related research is also lack. For narrowing the gap in detection technique between developed countries and our country and eliminating the Technical Barrier in the International Trade, it is more important to detect irradiated food.2-Alkylcyclobutanones (2-ACBs) mentioned in this paper,are the specific products of the irradiated lipid-containing food. The research results showed that the2-ACBs come from the fatty acids or triglyceride ester in food by irradiating. The concentration of2-ACBs is increasing linearly with the absorbed doses rising. Up to now, the2-ACBs only have been found in the irradiated food containing lipid, however, they have never been found in any non-irradiated food. So it can be used to as indicators to detect lipid-containing food irradiated or not, or to evaluate the absorbed dose.2-Dodecylcyclobutanone (2-DCB) is one kind of2-ACBs, which is the radiolysis product of palmitic acid and palm triglyceride in food. The alkyl chain of2-DCB is short and saturated, and2-DCB are more stable compared with other kinds of2-ACBs. In most food, the palmitic acid content is higher than most of other fatty acids, it is more practical to use its radiolytic product,2-DCB, as the main specific marker detecting irradiated food. The characteristics of2-DCB in irradiated food containing fat are the main objects of this paper. The quantitative analysis method have been studied and the pretreatment and instrumental analysis condition have been explored. A new method using of ultra-performance liquid chromatography-electrospray ionization tandem mass spectrometry (HPLC-ESI-MS/MS) for the analysis of2-Dodecylcyclobutanone (2-DCB) in irradiated foods containing fat has been firstly developed. The main projects and research results are as follows:1.It’s worth noting that a new ultra-performance liquid chromatography-electrospray ionization tandem mass spectrometry (HPLC-ESI-MS/MS) method for the analysis of2-Dodecylcyclobutanone (2-DCB) in irradiated foods containing fat has been firstly developed. Moreover, the conditions of chromatography and mass spectrometry were optimized.MS:electrospray (ESI) with multi-ion monitoring (MRM):positive ions. The results show that the best fragment of2-DCB is90voltage. The precursor ion is m/z239.1, the Qualitative ion is m/z94.8which has maximum ionic strength at15ev and the Quantitative ion is m/z108.8which has maximum ionic strength at10ev. The calibration curves were linear in the range of5-100μg/L for2-DCB.The method has the advantages including rapid analysis, high accuracy, good reproductivity and stability.2. By optimizing the type and concentration of eluting solvent, and the extent of deactivated neutral alumina on the extraction efficiency, the extraction condition of2-DCB was established as aether/hexane (1:99) and5%extent of deactivated neutral alumina were. It showed that the neutral alumina column separation method had lower cost,larger quantity of analytic sample, and high efficiency as compared with the EN1785method.
