Veterinary is an indispensible and important part in modern animal husbandry and aquaculture industry. The abuse of veterinary has led to high residue of it in the tissue of animals frequently, which has been attracting attention more and more attention. The relatively perfect analysis technique of liquid chromatography coupled to tandem mass spectrometry (LC-MS/MS) is based on the powerful isolation ability of HPLC and high sensitivity, selectivity and specificity of MS and it is widely used in the fields of veterinary residues analysisThis paper focus on the study on the application of HPLC-MS/MS in veterinary residues of high polar analysis, including the establishment of analytical method and the studied of matrix effect in HPLC-MS/MS.This paper includes three parts as follows:In chapter one, the background and the significance of this dissertation was elucidated. The basic theory of MS and MS/MS, the emergence, judgement and elimination of LC-MS/MS matrix effect was reviewed.In Chapter two, A sensitive liquid chromatography coupled to tandem mass spectrometry (LC-MS/MS) method was developed for the simultaneous determination of colistin and polymyxin B residues in animal derived foods. The target compound was calibrated with matrix matched standard solution, and thus eliminated the influence of matrix effect and the matrix effect of this method was studied by the way of mathematical statistics. The results shown that the established method was simple with high sensitivity and good reproducibility.In Chapter three, a sensitive LC-MS/MS method was developed for the simultaneous determination of Spectinomycin, Kanamycin, Amikacin, Gentamycin, Spectinomycin, Apramycin and Neomycin residues in animal derived food. The target compound was calibrated by matrix matched standard solution, the influence of matrix effect was eliminated and the matrix effect of this method was researched by the way of mathematical statistics. The simultaneous quantitative analysis and confirmation of the six aminoglycoside antibiotics residues in animal derived foods was realized.
