Iron is one of the most important elements for plants. There are many reports about absorption, transport and use of iron. Plant iron transport protein plays an important role in the situation when plants in the low iron stress resistance. There are not much reports about the regulation of transcription and post-translational. This research used the arabidopsis material of0x29/38witch is over excessive FIT and AtbHLH38genes in the wild type. We use this material to separate the new regulation genes for IRT1gene expression, and we are trying to fill in the blank of this area to consummate the regulation network of this gene.This study used the double excessive expression strain0x29/38to EMS mutagenesis. Use the phenotype of Ox29/38is not going to chlorosis when its in the iron deficiency conditions, we sowed the seeds of the next generation of mutagenic0x29/38in the1/2MS-Fe solid culture medium. We pick up the chlorosis plants under the iron deficiency conditions. This is the initial screening. Then we observed the phenotype and the consistency of phenotype in the next generation. Finally, we got four candidate mutant strains witch are0x29/38-20,0x29/38-29,0x29/38-31,0x29/38-2-4. We abserved the phenotype of the four candidate mutant and analysed the genes expression of them.Based on these data, we concluded that ox29/38-20and ox29/38-29were the target mutants we want to get, because the two mutants couldn’t accumulate the IRT1protein under iron sufficient condition.
