LFY gene plays an important role in floral development. As a floral meristem identity gene, LFY not only control the transition from vegetative to reproductive phase, but also function as upstream regulator of floral organs identity genes and is a key intergrator of the outputs of floral inductive pathways to induce floral meristem transformation. API, a downstream gene of LFY, is a MADS-box gene. MADS-box genes encoding a family of transcription factors and they control diverse developmental processes in flowering plants ranging from root to flower and fruit development.In this study, a MADS-box gene was isolated from Dendranthema lavandulifolium. The completed cDNA sequence is1029bp, encodes237amino acids and a terminating codon, named DAP1. The results of Blast showed that both had highly homologous with MADS-box genes, the analysis of conserved regions of DAP1indicated that it had a very conserved MADS-box region and a littler conserved region K-box region, which was identical with the typical structure of the MADS-box genes. The DAP1predicted protein is66.93%identical to API and NAP1-1.Real time PCR showed the expression of DFL and DFL after photoperiodic induction in Dendranthema lavandulifolium. The two gene’s expression was all rapidly high after short day treated. The tendency of DFL was ascended firstly and reached the first peak on the4th day, then descended. On the10th day it ascended again. Until the16th day, it reached a second peak, finally descended. The tendency of DAP I was ascended firstly and reached the first peak on the4th day, then contain a low level. By the result of expression, we could predict that DFL controls the expression of DAPI in the floral development.
