| Objective:The sleep activity of Drosophila melanogaster has biological rhythm,which is controlled by gene.in order to screen the genes,which is related to sleep.we have screened sleep-related genes.Methods:We choose the7-day old wild CS strains prosopon and the strains of Drosophila melanogaster with gene5601,8904,7061,7146,27327,669,8103,691,9697,24416,26525,5411,3096,5877,7682deletion were used in the research. Drosophila Activity Monitor System(DAMS) was used to record the Drosophila’s total sleep duration within24hours. The data was contrastively analyzed according to different kinds of strains.In finally,we confirm that the gene fragment of Df(3R)Esp13/TM6C affect the total sleep time,then we choose the Drosophila melanogaster of RNAi1、2、3...in this gene fragment and the7-day old wild CS strains prosopon as our research object, Drosophila Activity Monitor System (DAMS) was used to record the Drosophila’s total sleep duration within24hours.And then we are sure that which gene affect the total sleep time.Result:1、Compared with the prosopon of wild CS strains, the total sleep duration of5601strain was significantly shorter,P<0.05.2、We do repeated experitment in gene delect5601strain,it has Reproducibility, P<0.05.3、7061、8904、7146、27327、691、669、8103、5411、9697、24416、7682、26525、3096、5877strains had no significantly differences. P>0.05.4、Compared with the prosopon of wild CS strains,the total sleep duration of FBst0025845、FBst0031466、FBst0028712、FBst0035759strain of RNA interference were significantly extended.P<0.055、Compared with the prosopon of wild CS strains,the total sleep duration of FBst0025973、FBst0026321strain of RNA interference had no significantly differences, P>0.05.Conclusion: 1、The gene fragment of Df(3R)Esp13/TM6C reduced the total sleep duration.2、Nf1、Exo84、gro are related to sleep,which are wake-promoting genes.3、The gene fragment of Df(3R)Espl3/TM6C reduced the total sleep duration which is multiple genes that work together. |
