In this paper, we investigated the effects of pulse microwave with different irradiation parameters including pulse field intensity, pulse number and pulse duration, respectively on the newborn rat cardiomyocytes in vitro and the rats’ cardiac tissues in vivo. The apoptosis of the primary cultured newborn rat cardiomyocytes and the rats’ cardiac tissues is mainly taken into account. The dose-effect relationship between the pulse microwave irridiation and the biological effects from two levels, cardiomyocytes in vitro and cardiac tissues in vivo, has been figured out. And the threshold value of the pulse field intensity, which resulted in obvious biological effects has also been obtained.The biologicial effects of pulse microwave irradiation with different parameters on the newborn rat cardiomyocytes in vitro has been investigated by using flow cytometry. The biologicial effects of pulse microwave irradiation wi th different parameters on rats’ cardiac tissues in vivo has been figured out by a series experiments including the production of heart paraffin slice, observation under microscope, the statistics of hyperemia dots and TUNEL.The results showed that the trend of the biologicial effects on the newborn rat cardiomyocytes in vitro and the rats’ cardiac tissues in vivo is upwards in a certain range, with the increase of the pulse number or the pulse field intensity. The normal cell rate of cardiomyocytes decreased. While the apoptosis rate and the necrosis rate of cardiomyocytes increased. The apoptosis rate of the rats’ cardiac tissues increased. The number of hyperemia dots on the endometrial of left ventricular increased. While the counterparts on the adventitia remained steady. In summary, we obtained the threshold value of the pulse field intensity,6.18kV/m-7.02kV/m, which resulted in obvious biological effects of cardiomyocytes under our experiment conditions. The counterparts of cardiac tissues is7.00kV/m-8.05kV/m. The results provided important reference for the research of safe threshold on both cellular level and animal level.
