PrPCis a kind of neurons glycoprotein which expressed in cell membrane andhas the ability of neuroprotective. PrPSCexpresses in cytoplasm and has neurotoxiceffect on cell. There is no clear evidence about how PrPCturned to PrPSC. Therefore, itis necessary to research on other PrP-like protein.Protein Shadoo is a PrP-like protein that has wide exist in mammals even to fish.Recently, reaserch on PrP protein are focus on PrP, Doppel and Shadoo. Shadoo andPrP both expressed in CNS (central nervous system) and may possess overlappingfunction, that unlike Doppel which expressed in reproductive system. Therefore, thereaserch on function of Shadoo protein in CNS can contribute to study the function ofPrPCand how PrPCturn to PrPSC.Animal behavioral neuroscience experiments are the important characteristic toevaluate neural state. Behavioral neuroscience experiments which included evaluatethe function of protein, neural state, state of mind and so on. In this work, we useMorris water maze which most frequently used to evaluate learning and memoryability and open field as test instruments. The most standard general measure of motorfunction is spontaneous activity in the open field.Behavioral neuroscience experiments was used to study on Shadoooverexpression in mice, compared with wild type mice, and then the partialphysiological function of Shadoo in vivo was found. Immunohistochemistry andMALDI imaging was used to detect the location and expression level of Shadooprotein in brain. The change of brain tissue was analyzed with protein Shadoooverexpression. Two-dimensional electrophoresis was used to analyze other proteinexpression level in brain after Shadoo overexpression.In locus of place navigation test, compared with wild type mice, the latency oftransgenic mice were shorter on the4th,5th,6th,7th, especially on the4th(P<0.05). In the locus of probe trial test, compared with wild type mice, the time offirst crossings platform location was shorter in transgenic mice. The open field experiment detects the number of spontaneous activity in mice. The result is that themotor function of transgenic mice is higher than wild type mice. The number wassignificant difference (p<0.05) on1th,4th, and on2th,3th (p<0.01). The result ofWestern Blot shows that Shadoo expression level is higher than wild type in micebrain. The result of Immunohistochemistry and MALDI imaging showed that Shadooexpression level is significantly higher than wild type in the transgenic micecerebellum. The result of two-dimensional electrophoresis shows that total18proteinsare different in transgenic mice brain.According to above experiments, we finally conclude that Shadoooverexpression in mice can enhance the ability of learning and memory. Shadoooverexpression in mice can also enhance motor function and perform exiting-likebehavioral.
