Dynamic Light Scattering is one important branch of light scattering technologies,which can be used to reckoned diffusion coefficient of solution molecular or particleand fluid mechanics radius, and some dynamic mechanics parameters with thedetermination on slight frequency shift of light scattering and of which angulardependence; and which could determine the distribution of particle diameter with theaid of computers. With the advantages of low cost, quick survey speed, high precisionand no demand on samples, Dynamic Light Scattering has a broad application forvarious scientific researches. In biomedical, cancer is the enemy of life, which has longbeen a focus for concern. It is little to research on large molecules of cancer cells withDynamic Light Scattering, so this research is proposed in this paper.In this paper, in order to ensure the accuracy of the experiment calibratingļ¼Œthesolution of standard polystyrene latex spheres is used to calibrate the laser lightscattering instrument BI-200SM.Due to the different concentrations of the solution have certain impact on themeasurement results, so the different concentrations of bovine serum albumin solutionswere measured before the dynamic light scattering measurements of the intracellularprotein solutions. The results show that, within a certain concentration range, theautocorrelation function curve of photons intensity tends to stable smooth withincreasing concentration, while the delay time decreases and the relative error ofmeasuring particle size decreases. The experimental results show that in the dynamiclight scattering experiment, concentration of samples should not be too high or too low.To distinguish light scattering characteristics of protein between in hepatocyte andin HCC with the technology of Dynamic Light Scattering, the normal hepatocyte andHCC is cultivated, and then their protein is extracted and held differently; the extractionis measured with a wide-angle laser light scattering spectrometer; and then according tolight-intensity, particle size and particle size distribution data got from the experiment,this paper analyzed the different light scattering characteristics of protein between inhepatocyte and in HCC. This study helps people to promote the early diagnosis ofprimary liver cancer, and illustrates that dynamic light scattering is an effective meansof light scattering techniques in the domain of biological macromolecules.
