| Cells are the basic unit of biological tissue and cell activity detection is an important part of cell biology's experimental research. With the molecular processes deep research within living cell, it has already become an urgent request for technology to improve the accuracy of cell's activity detection based on the image.To resolve this key technical issue, we separate living neurons from mouse brain tissue, and then lock-in the cell which considered to have normal electro- physiological characteristics in the phase contrast microscope; we build a model of cell's activity vary through using mitochondrial function inhibitor(CCCP), Nystatin, Trypsin to induces neurons to death, meanwhile tracking and take the photo- graph in the same time interval and record digital microscopy image. After analyze the images, we find out that different apparent characteristics have different active states; In the Matlab programming environment, we used a programme which compiled by myself to analysis the images'contrast and equivalent gray lines which have the same gray-scale value in the edge of the cell image. The results of the programme demonstrated that the image of cell which active state strong relatively has large contrast value and has perfect equivalent gray-scale lines. The equivalent gray-scale lines in the edge are parallel each other approximately and similitude with the cell's shape. With the cell viability eliminate the equivalent gray-scale lines changed regularly along a specific direction.Contrast is a statistical characteristic values which based on the gradient of gray image of cells for statistical analysis and is the collective performance of all the pixels. Equivalent gray-scale line at the edge of cell's image is a result which were work out based on the interpolation calculate with pixel values nearby the location and is the pixels'collective performance which have the same gray-scale values. These two methods both can distinguish the images of cell which have different cell activity and have simple, fast characteristics, don't to distinguish the cells'shape and avoid the effects of staining and chemical examination.In conclusion, this study provides a method which can determine cell activity in the state of non-contact, non-chemical intervention and provides strong technical support for cell physiology, cell pharmacological in cell biology... |
