ObjectivesThe present study was carried out to investigate the functional status of genital system in pregnant women in e-waste region and developmental condition of their foetus and to explore the biological effect of noxious substance. We intend to offer baseline data and scientific evidences of our research to the other further studies.Methods1. Sample collecting and field investigationOne hundred pregnant women who delivered in F-hospital and G-hospital between January and July in 2006 were participated in this study with their written informed consents.Venous blood, umbilical core blood, placenta tissues and umbilical core tissues were collected for labtory analysis. The health information of the pregnant women and their spouse were also inquired after parturition.2. Methods and indexes of labtory analysisMDA, SOD, and GPX in blood and tissues were analyzed by spectrophotometric method. The levels of FSH, LH, and GH in blood were survey using radioactivity immunofluorescence. The placenta tissue and umbilical core tissue were collected to detect expressional abundance ofα-estrogen-receptor,β-estrogen-receptor and progesterone receptor by real-time PCR.3. Statistical analysisStatistical analysis was performed using SPSS 13.0 statistical software. Values were considered significantly different at the 0.05 level.Differences between two groups were tested using independent t-test,χ2–test and rank-sum test. The correlated relationships were analyzed by Pearson and Spearman rank correlation.Results1. Information of questionnaire1.1. Basic information of pregnant women In the study, the average age is 24.89±3.35 years old in the exposure-group, and 27.82±4.42 years old in the control-group;the average time of habitation is 19.77±10.10 years in the exposure-group, and 22.17±9.76 years in the control- group.1.2. Life conditions of pregnant womenRatio of drug history and no drug history in progestation, get touch with harmful physical factors and not get touch with harmful physical factors are no difference between two groups. All the pregnant women are not getting touch with harmful chemical factors, and no psychiatric history, family history of congenital anomaly and malignant neoplastic diseases.1.3. Average weight of neonatusThe average weight of neonatus is 3343.18±510.77g in exposure-group, and 3212.26±482.60g in control-group, there were no difference between two groups.1.4. Basic information of pregnant women's spouseStatistical analysis showed that there were no difference between two groups about getting touch with harmful physical factors, the history of smoking and drinking.2. Biological parameter results2.1. Results of oxidative stressThere were no difference about MDA between two groups'venous blood,the exposure-group's MDA in placenta tissue and umbilical core tissue was higher than control-group(P<0.05),but the exposure-group's MDA in umbilical core blood was lower than control-group(P<0.05).The exposure-group's SOD and GPX were lower than control-group in venous blood , umbilical core blood, and umbilical core tissue(P<0.05). The exposure-group's SOD in venous blood , and umbilical core blood higher than control-group(P<0.05).2.2. Level of hormoneThe level of FSH in venous serum was no difference between two groups, but the exposure-group was higher than the control group, the level of FSH in umbilical core blood was no difference among two groups.The level of LH and GH in venous serum was difference between two groups(P<0.05), the exposure-group was lower than the control group.2.3. Gene expressionThe gene expression of ERα, ERβand PGR in placenta tissue was different between two groups, the exposure-group was lower than the control group,decreasing 37.63%,90.75%,98.62%, and the gene expression of ERα, ERβand PGR in umbilical core tissue was different between two groups, increased by 294.85% of Eαand decreased by 57.43%,46.98% respectively with ERβand PGR.ConclusionsIn summary, the noxious substance in E-waste region can influence the human health by the oxidative stress and impact the level of hormone and the environment related gene expression.
