Objective:To detect potential proteins which may play a key role in pathogenesis andearly diagnosis for Acinetobacter baumannii sepsis, we compared the protein profile ofneutrophils of na ve rats with that of rats infected with Acinetobacter baumannii.Methods:24adult male Wistar rats were divided into normal control group,Acinetobacter baumannii sepsis in6hour group and Acinetobacter baumannii sepsis in12hours group, normal control group by intraperitoneal injection of saline,Acinetobacter baumannii sepsis group by intraperitoneal injection Acinetobacterbaumannii (ATCC19606), and Separation of neutrophils from peripheral blood after6hours and12hours, extraction of neutrophil total protein, using two-dimensional gelelectrophoresis separation of proteins and silver staining, using ImageMaster softwarefor analysis and comparison, some of the proteins were isolated and verified by linemass spectrometry analysis and Western blot.Results:1. Two-dimensional gel electrophoresis protein profiles of resolution andreproducibility are very well, Each group glue protein spots expressed are about2900,the average matching rates were95%,90%,91%.2. Fifty proteins from the neutrophils of rats with Acinetobacter baumannii sepsiswere found to be different from those of naive rats. Forty-one of these proteins wereable to be identified successfully using two-dimensional electrophoresis, they belong tothe antioxidant proteins, signaling proteins, cytoskeleton and regulatory proteins, energymetabolism and protease protein, in which24were up regulated, and17were downregulated3. Some proteins were selected to immunoblot validation, which from41proteins, theimmunoblot validation results are consistent with the two-dimensional gelelectrophoresis and mass spectrometry results.Conclusion:1. The protein profile of neutrophils in rats with Acinetobacter baumannii sepsis issignificantly different from that of naive rats,and the proteins of Acinetobacterbaumannii sepsis in6hours group and12hours group also have significantly difference. 2. In this research forty-one proteins were identified successfully, they belong to theantioxidant proteins, signaling proteins, cytoskeleton and regulatory proteins, energymetabolism and protease protein, Changes in these proteins were closely related toactivation or inhibition of neutrophils in Acinetobacter baumannii sepsis3. Antioxidant proteins and signal transduction proteins of differential proteins mayplay a key role in Bauman Acinetobacter sepsis, and may provides new theoretical inearly diagnosis and treatment in Bauman Acinetobacter sepsis.
