[Objective] Application of tow-dimensional gel electrophoresis followed by MALDI-TOF-TOF mass spectrometry to identify proteins differentially expressed in the lesion from patients with systemic lupus erythematosus (SLE)and the skin from normal control group. To explore the pathogenesis and some effective biomarkers of SLE.[Methods] Application of tow-dimensional gel electrophoresis, silver staining and technology of MALDI-TOF-TOF mass spectrometry to analysis the differential proteins expression in the lesion from45patients with SLE and the matched skin from normal control group.[Results]1,There are22differential proteins expressed. There were6differential proteins only expressed and14differential proteins highly expressed in the lesion of SLE. There were two proteins only expressed in the skin of normal control group.2, The results showed that10differentially expressed proteins were obtained with MALDI-TOF-TOF mass spectrometry. Eight proteins among them were searched in http://www.genecards.org.3,The expression of Heat shock70-kDa protein, Heat shock90-kDa protein1, SCCA1, SCCA2, Keratin2e, Keratin6and Involucrin proteins increased in the lesion of SLE, but the expression of Keratinl decreased. There were not proteins expression in the normal group.[Conclisions] There are some different proteins expresssion between SLE lesion and normal skin. The high expression of Heat shock protein (HSP) may assoicte with HSP antibody, SSA and SSB antibody. The high expression of Keratin2e,Keratin6, Involucrin proteins and the low expressionof Keratinl in lesion perhaps relate to keratinocyte activation, cytokine release, keratinocyte abnormal differentiation and the forming of SLE lesion.
