Expression And Relationship Of HINT1and SKP2Protein In Hepatocellular Carcinoma

Objective1.To investigate the expression of SKP2and HINT1in hepatocellular carcinoma and its relationship with clinicopathologic characteristics.2.To investigate the effects of HINT1on the proliferation, apoptosis and expressions of related SKP2, HINT1protern in hepatocellular carcinoma cells QGY-7701by Western-lotMethod1. The expression of SKP2and HINT1was evaluated in hepatocellular carcinoma tissues of40patients, liver cirrhosis tissues of20cases, and normal liver tissues of12patients by the immunohistochemical method。2. To observe the effects of HINT1on the proliferation, apoptosis and the expression of SKP2after and before HINT1was transfected into QGY-7701cells:(1) Cell culture (2) Four groups of cells were observed for their morphological changes under light microscopy.(3) Cationic liposome transfection method was employed to transfect HINT1into hepatocellular carcinoma cell line.The cells was divided four groups:①Control group: QGY7701.②:lipofectamine group:QGY7701+lipofectamine③pcDNA3.1(+) group: QGY7701+lipofectamine+pcDNA3.1(+)④INT1group:QGY7701+lipofectamine+pcDNA3.1(+)/HINT1(4) MTT method was employed to detect the proliferation of cells and draw the Growth curve (5) Western blot was used to observe the expression of HINT1and SKP2after and before HINT1was transfected into QGY-7701cells.Result The positive rate of SKP2(37.5%) in40case hepatocellular carcinoma was significantly higher than that in adjacent tumor tissues (5%) and normal liver tissues (negative expression) The expression of SKP2was closely correlated with differentiation (p<0.05),metastasis (p<0.05)。 but was not associated with age,gender, AFP level and HBsAg(+)。The expression of SKP2was Moderate negatively correlated with the expression of P27(r=-0.599, p<0.05)。HINT1can reduce the growth of QGY-7701cells。The expression of HINT1and SKP2in hepatocellular carcinoma cell lines varied after being transfeced HINT1gene.Conclusion:SKP2is highly expressed in hepatocellular carcinoma and the expression of Increase with the reduction of the degree of differentiation。HINT1gene may be related to the occurrence and development of hepatocellular carcinoma。 HINT1can significantly reduce the growth of the primary human hepatocellular Carcinoma cell。 HINT1Can significantly reduce the protein expression of SKP2。Combined detection of SKP2and HINT1index of clinical value for patients with hepatocellular carcinoma, diagnosis, prognostic evaluation and comprehensive treatment program designed to provide a strong theoretical basis。...
