Effect Of Yunnan Propolis On Tongue Cancer Cells In Vitro

[Objective] This thesis focused on the yunnan EEP in the role of the tongue cancer. To observe the EEP extract forces on theinfluence of ultrastructure and proliferation of cell. Preliminary discussion EEP extract may promote the mechanism of tongue cancer cell apoptosis,In order to provide the basis for action of propolis fight tongue cance.[Methods] Tongue cancer cell and oral normal mucosa fibroblasts were cultured in different concentrations of EEP which were diluted at25ug/ml,50ug/ml, lOOug/ml,200ug/mL,400ug/ml in vitro. Through the MTT method to detect the effects of EEP to the growth of tongue cancer cell and oral normal mucosa fibroblasts.Adopt inverted microscope and transmission electron micro-scope observe structure morphological chang of tongue cancer cell after different concentrations of EEP cultured different time. Expressions of apoptosis associated proteins, such as Bcl-2, Bax, and Caspase-3were analyzed by immunohistochemisty.[Results] EEP could inhibit proliferation of tongue cancer cell in vitro in a time-and dose-dependent manner. EEP could inhibit proliferation of normal mucosa fibroblasts in vitro in a time-and dose-dependent manner. But at the same time and at the same concentration EEP still significant difference role of two kinds of cells (p<0.01). Along with the propolis concentration increases and the extension of time, the tongue cancer cells to stick wall rate is lower, the more obvious cells form change,were observed under inverted microscope; tongue cancer cell apoptosis of scale to form change, the cytoplasm empty bubble increased, mitochondria swelling to go round, tiny hair cells myoglobin chromatin condensed,the nucleus shrinks and broken, chromatin the plaques edges together, mitochondria dissolved cristae formed much bubble body and apoptotic body,were observed under transmission electron microscope; In cell apoptosis related proteins, the results show that,Bcl-2expression was significantly down-regulated in response to EEP treatment whereas Bax and Caspase-3expressionswere up-regulated.[Conclusion] EEP can inhibit tongue cancer cell proliferation, propolis have cell toxicity to tongue cancer, it can cause people tongue cancer cells become pyknotic nuclei scale, chipped, nuclear chromatin the plaques form gathered, mitochondria swelling to go round and apoptotic body form; EEP can increase cell apoptosis in vitro, which may relate to the down-regulated of Bcl-2and up-regulated of Bax and Caspase-3.
