Objective To investigate the change of the microvascular density (MVD) of proximal bile duct after the bile duct of different plane was injured, In order to understand the change of blood supply in proximal bile duct and the correlation between the change of blood supply and the time after the bile duct of different plane was injured.Methods Clean and healthy forty-five SD rats were randomly divided into3groups (15each):control group, low bile duct skeletonized and injured group,high bile duct skeletonized and injured group, each group was equally divided into three subgroups according to the different time of observation, including3,7,14days subgroups.at3,7,14days after operation,five rats were killed, and the common proximal extrahepatic bile duct tissue was cut,then one of five specimens was observed after hematoxylin-eosin staining (HE), and expression of the MVD in proximal bile duct was determined by immunohistochemistry.Results HE shows:After the bile duct injured, with the extension of time, bile duct organization in new blood vessels increase gradually, bile duct cells have gradually degeneration, necrosis trend, bile duct tissue edema, loose increasingly obvious. Immunohistochemistry shows:after the bile duct injured, the MVD of proximal bile duct is higher, the higher the plane of bile duct is injured, the higher the MVD of proximal bile duct increase, the MVD of high bile duct skeletonized and injured group is higher than low bile duct skeletonized and injured group,(P <0.05), the MVD of low bile duct skeletonized and injured group is higher than the control group (P<0.05). After the bile duct injured, with the extension of time,the MVD is higher,14days is higher than7days and3days (P<0.05),7days is higher than3days (P<0.05).Conclusions After the bile duct injured, the MVD of proximal bile duct increased, the higher the MVD of proximal bile duct increase, and with the ischemia time delay, the more the MVD obvious near side bile duct increase; although the MVD increased after proximal bile duct injured, but the proximal bile duct still showed ischemic pathological changes. This result prompt that the proximal bile duct ischemia is universal after bile duct injury, the ischemia of the high bile duct skeletonized and injured is more serious than the low bile duct skeletonized and injured, with the time delay the ischemia is more serious,but the increased new blood vessels couldn't satisfy the bile duct blood and oxygen supply.
