Objective To explore the effects of Transforming Growth Factor-beta1(TGF—β1) on repair of dog with bile duct injury at different locationMethods 25 Beagle dogs were randomly divided into normal control group,bile duct injury group,superior-position injury repair group,low-position injury repair group,mid-position injury repair group.The model of bile duct injury was constructed and bile duct injury was repaired on different position.All dogs were sacrificed on the 15thday of post-operation and we took all collections of bile duct. The general state of all dogs and postoperative complications were followed up.All collections were carried out with HE staining to observe and Streptavidin Peroxidase Method to detect expression of TGF-β1 on bile ducts.Results Light microscopy showed that mucous membrane exuviated,arrangement of collagen fibers was tangle,new vessels increasing and vessel wall thicking in bile ducts of injury repair groups.Immunohistochemistry showed that expression of TGF—β1 in bile ducts was negative in control group and was hadro-masculine in injury repair groups,which indicated that significant difference existed on different positions(P<0.05);Significant difference existed on low-position relative to mid-position and superior-position(P<0.05)and no statistical significant existed on mid-position relative to superior-position(P>0.05)about expression of TGF—β1 in bile ducts in injury group.Comparison about expression of TGF—β1 in the same position of all groups:Significant difference existed on control group relative to injury repair groups and injury group(P<0.05)and no statistical significant existed on injury repair groups relative to injury group(P>0.05)in these low-positions. Significant difference existed on injury repair groups relative to injury group and control group(P<0.05)and no statistical significant existed on control group relative to injury group(P>0.05)in these mid-positions and superior-positions.Conclusions The quantity of expression of TGF—β1 in bile ducts of injury repair increased and was different on different positions,which was relatively more on near to duodenum(low-position)than on near to porta hepatic(superior-position).In the same background of BDI,the expression of TGF—β1 on mid-position and superior-position was less than that on low-position.Because of the direct correlation between scar accrementition after repair and restenosis of bile duct after postoperation, the incidence rate of restenosis in superior-position bile duct-jejunum anastomosis than that in low-position.
