[Background] Mitogen-activated protien kinase kinase4(MKK4) is an important signal transduction protein in cells, it can active and transfer a variety of extracellular signals into the nucleus by phosphorylate them, and activate a variety of nuclear transcription factors, participate in cell growth, development, division and differentiation, etc. Research shows that MKK4is related to cancer occurrence, metastatic development, resistance and prognosis.This experiment used HRP immunohistochemical method to detect the expression of MKK4protein in osteosarcoma, bone giant cell tumors, osteochondroma, than discusses the relationship between MKK4protein and malignancy of bone tumors, metastasis and prognosis of osteosarcoma.[Materials and methods]30cases of osteosarcoma,28cases of giant cell tumor of bone,30cases of osteochondroma were collected from the department of the third affiliated hospital of KuMing medical college, the tumor hospital of YunNan province, which were saved by paraffin after formalin fixed and decalcify by Rapid·Immuno demineralization liquid. HE dyed and Horseradish Peroxidase (Horseradish Peroxidase, HRP) immunohistochemical method were used in the specimens respectively, which were slice4μm continuousiy in every specimen. The known positive pathological of MKK4for was choosen as a positive control, with PBS instead of first antibody for the negative control. The positive expression of MKK4protien is yellow or yellowish-brown dyeing in the nucleus and/or cytoplasm. Calculate the quantity of positive cells at5vision of higher magnification in each section by The IRS rating scoring system. Analysis the data by SPSS statistics software.[Result]The expression of MKK4in osteosarcoma is higher than the bone giant cell tumor and osteochondroma; the MKK4is obviously expressed in both nucleus and cytoplasm in osteosarcoma; obviously expressed in cytoplasm, partially expressed in nucleus in the bone giant cell tumor; partly expressed in cytoplasm, rarely expressed in nucleus in osteochondroma. The IRS score shows that:the expression of MKK4in the cytoplasm and nuclear of osteosarcoma is obviously higher than the bone giant cell and osteochondroma; and is higher expressed in pulmonary metastasis than no pulmonary metastasis in in the cytoplasm; the expression in nucleus is in proportion to the malignant, metastasis and prognosis of osteosarcoma.[Conclusions] The MKK4is highly expressed in the nucleus andr cytoplasm of osteosarcoma, with a statistically significant difference, and is in proportion to the malignant, suggest that MKK4play a role in promoting osteosarcoma and the differentation in the bone, and is related to the metastasis and prognosis of osteosarcoma, can be used as one of a prognostic indicator of judgment osteosarcoma...
