Diabetes mellitus (DM) is a metabolic disease characterized by hyperglycemia.Itsincidence rate goes up these years. Its complications of large vessels and capillaryvessels always cause death and mutilation. Kidney and cadiovascular tissues are thepredominant target organs of diabetic chronic complications.We have proved that there are regional renin-angiotensin systerm in kidney, cardiacmuscle and smooth muscle of blood vessels. When the blood glucose goesup,renin-angiotensin system can be activated by it.This may accelerate the progress of thediabetic chronic complication.Angiotensin Ⅱ(AngⅡ)is the main active factor in therenin-angiotensin system. Its step-up indicate the activation of renin-angiotensin system.The step-up of angiotensin Ⅱ can cause abnormality of endothelium,constitutionremodeling and inflammations in blood vessels.These may greatly relate to diabeticnephropathy, left ventricular hypertrophy and atherosclerosis. Now, angiotensin Ⅱisconsiderd to be hormone with several effects. It can influence the proliferation, apoptosis,migration, Inflammation,fiberations of cells and ect.These effects are mainly through itscombination with angiotensin Ⅱ typer1receptor(AT1R). Recent studies suggest that theinhibition of renin-angiotensin system can proetct kidney and cadiovascular tissues indiabetes.The pathological basis of renal and cadiovascular disease in diabetes is disability ofcirculation.According to chinese medical science, qi-yin asthenia and obstruction ofcollaterals by blood stasis are the most important pathomechanisms in diabetic circulationdisability. Jin Li-da granules can invigorate vital energy,nourish yin, enlarge bloodvessels and dredge collaterals.In the clinic, Jin Li-da granules can protect kidney andcadiovascular tissues in diabetes. But we do not understand its mechanisms.Objective: This experiment is to observe the effects of Jin Li-da granules on the levels ofRAS in kidney,heart and abdominal aorta of experimental diabetic rats, so as to explorethe possible mechanism by which Jin Li-da granules protect kidney and cadiovasculartissues.Methods: We construct rats models of diabetic chronic vascular complications. Therats models were randomized into2groups: one group were cheated with Jin Li-dagranules while the other were cheated with placebo. After the interference, we measued the body mass, glucose,24H unrine protein, idney mass, heart mass; AngiotensinⅠ(AngⅠ) and Angiotensin Ⅱ(AngⅡ); the protein expression of Angiotensin Ⅱtype1receptor(AT1R) and Angiotensin Ⅱtype2receptor (AT2R) in kidney, heart andabdominal aorta of all rats.Results: Compared with Normal contronl group(CON) group, the kidney mass/bodymass(KM/BM), heart mass/body mass(HM/BM), blood glucose(BG),24-hoururine proteins (24h UP) in the Diabetic model group(DM) group significantly increased(P<0.05); the BM significantly decreased(P<0.05); the expression of AngⅠ,AngⅡ, AT1R and AT2R in kidney, heart and abdominal aorta were significantlyhigher(P<0.05). Compared with DM group, the KM/BM,24h UP in Jin Li-dagranules treatment group (DM+JLG) group significantly decreased (P<0.05), the BMsignificantly increased (P<0.05), and the expression of AngⅠ, AngⅡ, AT1R andAT2R in kidney, heart and abdominal aorta were significantly lower (P<0.05).Conclusion: The level of RAS in the kidney, heart and abdominal aorta of diabetic ratssupressed after that they are treated with Jin Li-da granules. And Jin Li-da granulesprobably be beneficial to protect the kidney and cardiovascular system in diabetic humans.
