Objective:To study the expression of Toll-like receptor4(TLR4), osteopontin in renal tubular epithelial cells exposed to high glucose. the effect of Exendin-4on the expression of TLR4, OPN in renal tubular epithelia cells exposed to high glucose, and provide scientific basis for GLP-1analogues Exendin-4fall blood sugar effect outside of the kidney protection.Methods:In vitro renal tubular epithelial cells (HK-2) were cultured with different concent-rations of Exendin-4the cell proliferation was examined by MTT colorimetry to determine the experimental dose of Exendin-4. randomly exposed to DMEM culture solution with low glucose (5.5mmol/L), high glucose(30mmol/L),10pmol/L Exendin-4intervention group,20pmol/L Exendin-4intervention group, or40pmol/L Exendin-4intervention group. After24h,48h and72h, Immunohistochemistry and RealTime-PCR used to determine TLR4,OPN protein and mRNA expression. The differences of data among groups were compared by One-Way ANOVA TestResult:1. MTT colorimetry suggest that0.1pmol/L,1.0pmol/L,10pmol/L,50pmol/L, dose of Exendin-4, cell proliferation compare with matched group was express no difference (p>0.05)100pmol/L dose of Exendin-4, cell proliferation was higher than that in the matched group(p<0.05), when200pmol/L,1000pmol/L dose of Exendin-4cell proliferation Significantly higher than that in the matched group (p<0.05)2. RealTime-PCR suggest that the expression of TLR4,OPN mRNA in the low glucose group lower. The expression of TLR4,OPN mRNA in the high glucose group increase as compared with the low glucose group, and the expression of time-dependent manner.The TLR4,OPN mRNA expression in the Exendin-4treatment group was significantly lower than that in the high glucose group, although it was higher than that in the low glucose group. Exendin-4markedly inhibited the expression of dose-dependent manner.3.1mmunohistochemistry suggest that The expression of TLR4,OPN protein in the low glucose group lower. The expression of TLR4,OPN protein in the high glucose group increase as compared with the low glucose group, and the expression of time-dependent manner.The TLR4,OPN protein expression in the Exendin-4treatment group was significantly lower than that in the high glucose group, although it was higher than that in the low glucose group.Exendin-4markedly inhibited the expression of dose-dependent manner.Conclusion:1. High glucose triggers an increase in the expression of TLR4and OPN in HK-2cells2. Exendin-4to the kidneys have certain protective effect may be by inhibiting TLR4, the expression of OPN, thus reducing the immune inflammation the damage to the kidneys.3. The reduce the role of Exendin-4on TLR4,OPN is the dose-depende.
