Objective:Type-2 diabetes is very common and frequently in today's society,and as risk of coronary heart disease,which can increase the incidence of cardiovascular events significantly. Fat metabolism disorder is one of the risk factors of atherosclerosis in type-2 diabetes cases. Abnormal increase of lipid deposite in cells and blood vessel walls, forming plaque, resulting in vascular stenosis, or occlusion. Therefore,the anti-atherosclerosis drug application,has a very important significance for prevention and treatment of heart diseases.The study takes SD rats which has similar AS formation with human being as example,to observe the expression of CD40,MMP-9,AP-1 on the arterial vessel wall and the relationship of Inflammation and plaque instability. The objective is to identify the effects of Rosuvastatin on early atherosclerosis in male rats with type-2 Diabetes Mellitus.Methods:Forty-five male SD rats were equally randomized to three groups.Group A was the rats with normal control(Group NC).Group B was the rats with Type-2 diabetes mellitus (Group DM).Group C was the rats with DM intervened with rosuvastatin(Group DR).There were 15 rats for each group.Group B and C were feeded with high-fat and high-sugar food.One month later,the rats were shot sigle dose of 25mg/kgSTZ intraperitoneal injection in Group B and Group C.Group A was shot citrate buffer.Group Cwas taken intragastric administration with 10mg/kg.d rosuvastatin.The intragastric administration was done 2 times per day at moring and night.Group A and B were taken intragastric administration with saline as control. After 16 weeks,Fasting blood tests the TC,TG,LDL. HDL and BG in all groups,and Immunohistochemistry tests Aortic wall CD40,MMP-9 and AP-1.Results:Compared with the control group,the level of TC,TG,LDL in blood were increased significantly(P<0.01)in Group B and Group C.HDL Significantly increased Compared with the control group (t=3.2863,9.6394,8.0155.8.1916,P<0.05).The expression of CD40,MMP-9,AP-1 in type 2 diabetic group, rosuvastatin group had significant differences compared with the control group(F=91.92~961.84,4=9.48~59.35, P<0.01) The CD40,MMP-9,AP-1 of Group C was significantly lower than those in Group B(F=36.86~716.82, q=8.59~37.86,P<0.05).The endothelial damage of the aortas in rosuvastatin group was less severe than in diabetes mellitus group.Conclusion:rosuvastatin can prevent early atherogenesis through alleviating the damage to the arterial wall by inhibiting the CD40,MMP-9,AP-1 expression and the monocytes infilitrating into the arterial wall.
