BackgroundTethered cord syndrome (TCS) is commen in Congenital spinal cord disease has been defined as a syndrome with a series of clinical symptoms and signs resulting in an abnormal increase of spinal cord tension due to pressure in termination of conus medullaris caused by various congenital or acquired factors. Nerve cells has been confirmed can be separation culture from central nervous system, but filum terminale, which located in termination of spinal cord has been considered as fibrous bands, which contains no neurons, and systematic researches about nerve cells in filum terminale were few. Our research attempt to separation culture filum terminale of children with TCS, observe status of nerve cells in flium terminals of children. Transcription factor SOX2, one key members of isoforms B of the SOX family, is an important gene in maintain stem cell characteristics. Recent research showed that over expression of SOX2are implicated in several cancers including lung cancer, esophageal cancer, gastric cancer, encephalophyma. There have been a lot of clinical researches on TCS at home and abroad, factor levels research has not been reported. This article initially investigate expressions of SOX-2in filum terminale of TCS patients and its influence in development in TCS. Objective1. Primary culture filum terminale of children with TCS, observe status of nerve cells in flium terminals, providing a new recourses of nerve cells.2. Evaluate expressions of Sox-2in both normal and abnormal filum terminale, investigate the possible mechanisms in progress of TCS.Materials and methods1. Filum terminale of children with TCS(donated by Third Affiliated Hospital of Zhengzhou University) was primarily cultured, the growth of the cells was observed under the phase-contrast microscope and all the cells were appraised by immunofluorescence.2.70children with TCS hospitalize in the Third Affiliated Hospital of Zhengzhou with10normal fulim terminale obtained by repair surgery of pure meningocele were included in the study, RT-PCR was used to detect the expression of sox-2. SPSS11.0was used for statistical analysis.Result1. Nerve cells as neuron, neuroglial cell(mainly neuroglial cell) can be cultivated from the fulim terminale of TCS children, which proves that nerve cells exist in fulim terminale.2. Compared with the low expressions of Sox-2in normal fulim terminale,the expression of Sox-2obviously increased in fulim terminale of TCS patient, and there were significant differences in specificity (P<0.05).Conclusion1. Under specified condition, nerve cells can be cultivated from the fulim terminale of TCS patient, which provide a new recourses of nerve cells, have certain degree of clinical significance. 2.
