AIM:To discuss the influence of lentiviral vector carrying the light gene induce gastric cancer cells apoptosis.Methods:Using lentivirus mediate Light gene infects SGC-7901 gastric cancer cells. The cells were divided into three groups:the experimental group, the negative control group and the blank group. Separately infected by obtained lentiviral particles, Lentiviral particles and PBS, the expression of Light was observed by RT-PCR and Elisa. The apoptosis of gastric cells was detected by flow cytometer.Result:The expression of green fluorescent protein was found in SGC-7901 cells infected with lentivirus 48h later, and cells infected with efficiency greater more than 80% from observed by fluorescence microscopy, slow lentivirus infected SGC-7901 cells efficiency was very notable. The result of RT-PCR showed that the electrophoresis of experimental group cells can be seen notable band at 128 base pairs. The result of Elisa showed that the protein concentration were no statistically significant differences among the three groups on the first day. But the statistical differences were significant after the first day. The result of flow cytometer showed that apoptosis of the experimental group cells was significant.Conclusion:Lentiviral vector carrying Light gene has been constructed, product hight recombined lenti viral virus. Lentiviral virus infected SGC-7901 cells and the Light protein could be expressed. Light genes mediated by lentivirus may be useful for inhibiting growth of gastric cells, and can induce gastric cancer cells apoptosis. This will facilitate the following exploratory development of light gene carried by lentiviral virus treat gastric carcinoma.
