ObjectiveTo observe the impact of estrogen(E2) on the proliferation and apoptosis of thyroid primary cells, and the impact on the apoptosis induced by DMBA.MethodPrimary cultures were isolated from 1 female benign thyroid tumor patient, 1 male benign thyroid tumor patient, and 1 malignant thyroid tumor patient, both tumor tissue and normal thyroid tissue, which were then classified into 6 groups. They were then grown with 10-11, 10-10, 10-9 or 10-8E2, and counted at 72h, with a bar made for each concentration. Some other cells were grown in 96-well plate with 10-11, 10-10, 10-9 or 10-8E2, the absorbances were gauged at 24h,48h,72h under MTT-test, with charts made for each concentration. And the left cells were grown in 96-well plate as same as above, with DMBA10ug per well for 0h,4h,8h,24h or 48h, also with charts made for each concentration. The data was analyzed by SPSS.ResultAdvancement on proliferation were observed both by cell counting and MTT on all 6 groups of cells, with P<0.05 when compared with the control group, but the impact declined when the concentration was too high. The apoptosis of the 5 groups induced by DMBA were inhibited, with P<0.05 when compared with the control group, but the impact declined when the concentration was too high.ConclusionE2 do press promoting effect on thyroid normal cells and tumor cells proliferation, but weaker when the concentration is higher than 10-8M. E2 do inhibit apoptosis of both normal thyroid cells and benign tumor cells, but weaker when the concentration is higher than 10-8M.
