| Objective: To establish an animal model of modified two-cuff technique orthotopic of liver transplantion in rat.Methods: 70 cases of rat liver transplantation had been established with modified cuff technique.The anastomoses of surahepatic vena cava and the perioperative treatment.Result: Among the latter 70 cases, the mean time of anhepatic period was 15 minutes, and mean operation time was minutes. The achievement ratio of operation was 96% and 2weeks survival was 90%.Conclusions: The modified two-cuffed methods is convenient and easy to duplicated. It can enhance the stability and survival rate of rat orthotopic liver transplantation models.PARTⅡ: The protective effect of edaravone on liver graft against IRI in rat OLT modelObjective: To study the neuroprotective mechanism of edaravone on rat brain damage following cerebral ischemia-reperfusion.Methods: 20 Couples of male rats, weighed 180-250g,were randomized into three groups. Perfusing and preserving solution were edaravone with wanwen in experimental group and wanwen solution without edaravone in the control group. The bile output was observed in two groups of liver grafts after portal vein reperfusion. Twelve hours after reperfusion, the recipients were sacrificed, serum ALT,AST were measured, apoptosis of the hepatocytes was analyzed by TUNEL and TNF-α,NF-κB,levels were detected by immunohistochemistry.Result: After 12 hours. The bile output in the experimental group was significantly higher than in control group. Serum ALT,AST levels in the experimental group were significantly lower than those in control group. In electron microscopy, the hepatocytes injury in the control group were significantly serious in the experimental group. In TUNEL and FCM analysis, the number of apoptosis in the hepatocytes in the control group was markedly increased compared with the experimental group. Immounohistochemistry study showed the TNF-α,NF-κB level in control group was significantly lower than in experimental group .Conclusions: 1.Edaravone can decrease ischemia-reperfusion injury in rat liver transplantation.2.The protective mechanism of Edaravone in rat liver transplantation may be:①R estraining TNF-αand NF-κB expression.②inhibition of janus kinase/signal transducer and activator of transcription (JAK/STAT) pathway on ischemia-reperfusion . |
