Objective Activation of the alternative pathway of the complement system, in which factor H (fH; complement fH [CFH]) is a key regulatory component, has been suggested as a link between obesity and metabolic disorders. To investigate the levels of serum Complement factor H,C reaction-protein in type 2 diabetes and to explore their roles in this disease and find the correlation between these adipokine hormone levels and the metabolic of glucose and lipids.Methods According to the body mass index(BMI),60 type2 diabetes patients were divided into two groups,one group was non-obese diabetes patients with BMI<25kg/m2 (28cases) and the other group was obese diabetes patients with BMI≥25kg/m2(32cases).There were 30 healthy persons in the control group.ELISA technique were employed to determine the serum Complement factor H, hs-C reaction protein were measured by immunoturbidimetric assay,HBAlc were measured by automatic saccharification analyzer;blood pressure,height,other data including body weight,waist circumference and hip circumference were measured to calculate BMI and waist-hip ratio(WHR).at the same time automatic analytical instrument was used to determine blood glucose and blood fat;insulin resistance index and insulin sensitive index were calculated by the HOMA-IR.ResultsThe levels of serum CFH and hs-CRP were increased significantly in obese T2DM group compared with that in control group(p<0.05); moreover, in obese T2DM group were increased compared with that non-obese T2DM group(p<0.05).Blood fasting glucose in patients with obese T2DM(9.9±3.9mmol/l) increased significantly (p<0.05) compared with the control group.so did insulin levels(12.9±5.6mIU/l) compared with the control(P<0.05).Correlate analysis showed that fasting serum CFH concentration was a positive correlated with BMI.HOMA-IR, fasting insulin,HBAlc and triglyceridemia(r=0.398,p<0.01;r=0.334,p<0.01;r=0.603,p<0.01;r=0.457,p<0. 01;r=0.313,p<0.05),also a negative correlated with HDL.There was no correlation between serum CFH concentration and blood pressure.ConclusionThe levels of serum CFH were increased significantly in obese T2DM group compared with that in control group and non-obese T2DM group.and correlate analysis showed that serum CFH concentration was a positive correlated with BMI,HOMA-IR, moreover a negative correlated with HDL, it predicted that serum CFH maybe was the link between obese and diabetes.The level of plasma hs-CRP were increased significantly in obese T2DM group compared with that in control group, and correlated positively with plasma CFH showed that hs-CRP and CFH were factor to predict obese developing to diabetes. |
