Objective To detect the expression of SDF-1, CD34, MVD at the PingPong Racket flap during the survival process, and investigate the role of SDF-1 at flap neovascularization .Methods The PingPong Racket flap, which pedicle's length breadth ratio were 4:2, each took an area of 2cm×2cm(group A),3cm×3cm(group B),4cm×4cm(group C),5cm×5cm(group D),6cm×6cm(group E), were designed and elevated on each five pigs' double dorsum . The flaps were served as the hypoxia ischemia flaps model. The survival area, histologic analysis, SDF-1 and CD34 enzyme-linked immunosorbent assay try at the distal flap were evaluated at days 0, 3, 5, 7, and 14 after operation respectively.Results①The living process and pathologic process of traditional flap and slender narrow pedicle flap were consistent. It could not postpone the flap living process that the flap pedicle became long and narrow;②When the ratio of the length to width of the slender narrow pedicle was constant, along with the flap area increased, the flap survival area also increased, but when the flap reached a certain area , the distal flap would necrosis;③In the same group of flaps, when time-lapse, the expression of SDF-1, CD34 and MVD increased, reached the plateau level after 5 days of SDF-1 ( A124.80±4.05, B137.85±3.03 , C166.53±2.98 , D72.80±2.63 , E 62.79±2.20 ), 7days of CD34 ( A16.76±0.62 , B17.60±0.72, C18.48±0.55 , D12.70±0.60 , E11.51±0.70 ), 7days of MVD(A 52.45±2.78 , B59.34±3.12 , C61.14±3.35 , D25.25±3.78 , E 24.46±7.46) , and then decreased gradually;④In the different groups of flaps, when the flap area increased , the expression of SDF-1, CD34 and MVD increased , and decreased at the area of 5cm×5cm , the flaps were partial necrosis. Conclusion①SDF-1, CD34, MVD play important role in the survival process of the PingPong Racket flap;②CD34 and MVD could indicate the neovascularization of the flap;③SDF-1 may play an adjusted role in the survival process of the flap.
