Cells between the fourth and tenth passages were used in this study. The reverse transcription and polymerase chain reaction (RT-PCR) method was applied to examine the production of the MMP-1,MMP-2,MMP-3 mRNA by demal fibrblast cells with SPC. Western blotting analysis was investigated the expression of the SPC-induced MMPs in human demal fibroblast cells in course of culture (2 to 48 hours).Results: Human demal fibroblast cells were successfully cultivated from foreskin digested with Dispaseā…”.The results of RT-PCR method revealed that SPC could increase the level of MMP-1,MMP-3 mRNA expression. The level begin to increase on hour 2 and persist throughout the entire culture period. But the increase of MMP-2 is insignificant. Western blotting analysis showed that the SPC-induced MMP-1,MMP-3 increased effect displayed time-dependent manner.Conclusion: In summary, these results suggest that SPC could have the potential to stimulate the production of elevated levels of MMP-1,MMP-3 in human demal fibroblast cells and balance ECM. Maybe one of the mechanisms that SPC improve skin wound healing.
