Cerebellar cells of newborn rats were used in this study,investigate the effects of10-HDA on the cerebellar neurons.Determine the best conditions of primarycerebellar neurons cultured in vitro,the suitable time of adding cytarabine to thecells,Different dosages of 10-HDA were added to the cultured cerebellar neuronswith final concentrations of 0.1μmol/L,0.5μmol/L,1.0μmol/L,5.0μmol/L,10.0μmol/L.Cell proliferation were detected by BrdU,NSE methods.Cell viabilitywas detected by MTT methods.Results were that:1.Cerebellum were isolated from newborn rats within 48h in the sterile conditions,meninges were stripped and cerebellum were degisted with trypsin,cell suspensionwere geted.Viable cells were up to 97% by trypan blue staining.The best plantingdensity was 1×106个/mL,the convert phase microscope was used to observe cells,cultured neurons were stick to the wall of the cultivation plate,cells were satiety.Cytarabine were added to the cells after planting 72h,and effecting 48h,theproportion of neurons were more than 92%.2.Brdu staining indicated that different concentrations of 10-HDA effected the proliferation of cerebellar cells,proliferation rate were that,the control group (6.8±0.83)%,0.1μmol/L group (9.8±0.84)%,0.5μmol/L group(13.4±0.97)%,1.0μmol /L group (15.2±1.31)%,5.0μmol/L group(9.2±1.30)%,10.0μmol/L group (7.3±0.91)%.0.1μmol/L group,0.5μmol/L group and 1.0μmol /L group were significantly increased than the control group( (p<0.05,p<0.01,p <0.01),5.0μmol/L group and 10.0μmol/L group had no significance compared with the control group,NSE immunocyto-chemisty showed that 0.1μmol/L group,0.5μmol/L group and 1.0μmol /L group were significantly increased than the control group( (p<0.05,p <0.01,p <0.01).3.MTT results showed that the OD values of 0.1μmol / L group,0.5μmol/L groupand 1.0μmol/Lgroup compared with the control group were significantly higher (p <0.05,p <0.01,p <0.01),the OD value of 5.0μmol/l group and 10.0μmol/lgroup had no significant difference compared with the control group.Conclusion:10-HDA effected the proliferation and survival of primarycultured cerebellar cells,and were related to the concentration of 10-HDA....
