Objective:To observe the phosphatidylinositol-3 kinase and protein kinase B (PI3K/AKT) signaling pathway in early osteoarthritis of the knee articular cartilage expression and to explore bidentata saponins on rabbit knee cartilage cells and the PI3K/AKT pathway.Methods:30 rabbits were randomly divided into 3 groups of 10 each, the normal group, model group and bidentata saponins group. Besides the normal group, osteoarthritis model were made by Hulth method. Morphological changes in cartilage were observed after Masson staining and marked by Mankin. No in synovial fluid was detected and chondrocyte apoptosis were measured by Flow cytometry,than expression of PI3K/AKT pathway in cartilage was disclosed by western blot.Results:1.The results of pathology change in articular cartilage:In gross specimen observation, the model group losed their luster cartilage and brightness with the gray surface. Model group, the cartilage thickness, cartilage cells arranged disorder.In light microscope, the cartilage become thicker and cartilage cells array disordered.2. The results of Mankin score:The scores of model group and bidentata saponins group were significantly higher than the normal group (P<0.05). Compared with model group, Bidentata saponins group were significantly lower(P<0.05).3.NO content in synovial fluid: The level of synovial fluid NO in model group was higher than normal group while the Bidentata saponins group was significantly lower than the model group (P<0.05).4. Detection of chondrocyte apoptosis:Chondrocyte apoptosis in nomal group showed a significant downtrend than the normal group(P<0.05).5. AKT expression in cartilage:The AKT protein expression in model group was higer than the normal group, the expression of Achyranthes bidentata saponins group close to the normal group while Bidentata saponins expression group close to the normal group.Conclusion:Bidentata saponins can reduce the NO content in synovial fluid and apoptosis level of cartilage cells. Furthermore, AKT expression in the rabbit knee was increased. The mechanism may relate to interfere with PI3K/AKT signal transduction pathway and improve the level of phosphorylation of AKT which was able to prevent and treatment the early osteoarthritis.
