| Objective To investigate the effects of prostaglandin E1 on expression of inflammatory in model with lumbar intervertebral disc herniation. To reveal the mechanism that prostaglandin E1 treatmented the lumbar intervertebral disc herniation in order to provide theoretical basics of the clinical treatment. Methods 60 male rabbits were randonmized into five groups: control group, sham group, 0.9% sodium chloride group, dexamethasone group and prostaglandin E1 group. 12 rabbits each group. The control group was not modeled, the sham group removed part of the lamina , the other groups were modeled . Four rabbits of each group were surgeried in the 7th day for 100mg tissue aroud the left lever root of L5-6 Disc. To inject preoperative anesthetics with 1% ketamine 40mg/kg by the ear vein slowly. The tissue was detected the lever of interleukin 1 (IL-1), nitric oxide (NO) and histamine (HA), to prove successful model. After the modeling , the control group and the sham group were not administered . Each rabbit of the 0.9% sodium chloride group was injected 0.9% sodium chloride 20ml by the ear vein slowly. Each rabbit of the dexamethasone group was injected 0.9% sodium chloride 20ml add dexamethasone 1mg/kg by the ear vein slowly .Each rabbit of the prostaglandin E1 group was injected 0.9% sodium chloride 20ml add prostaglandin E10.2μg/kg by the ear vein slowly. It was once every day, it was continuous for 10 days . In the 17th day (treatment for 10 days), four rabbits of each group were detected the lever of interleukin 1 (IL-1), nitric oxide (NO) and histamine (HA) with the same methods. In the 27th day (withdrawal for 10 days), four rabbits of each group were detected the lever of interleukin 1 (IL-1), nitric oxide (NO) and histamine (HA) with the same methods. Result In the 7th day after modeling, it was not significantly different that the lever of interleukin 1, nitric oxide and histamine of the tissue aroud the left never root of L5-6 disc, as compared with the sham group and the control group(P≥0.05). It was higher, as the 0.9% sodium chloride group, the dexamethasone group and the prostaglandin E1 group compared with the sham group, the control group respectively (P<0.05). In the 17th day (treatment for 10 days) and the 27th day(withdrawal for 10 days), it was higher, as the 0.9% sodium chloride group, the dexamethasone group and the prostaglandin E1 group compared with the sham group, the control group respectively (P<0.05). It was lower, the lever of interleukin 1, nitric oxide and histamine of the tissue aroud the left lever root of L5-6 Disc, As the dexamethasone group, the prostaglandin E1 group compared with the 0.9% sodium chloride group respectively(P<0.05). It was lower, as the prostaglandin E1 group compared with the dexamethasone group(P<0.05). Conclusion 1. It was successful on the models with lumbar intervertebral disc herniation . 2. Prostaglandin E1 could decline the lever of interleukin 1, nitric oxide and histamine that the tissue aroud the lever root with lumbar intervertebral disc herniation. 3.The mechanism that prostaglandin E1 treatmented the lumbar intervertebral disc herniation, which related to decline the levers of interleukin 1, nitric oxide, histamine of the tissue aroud the lever root. |
