ObjectiveBreast cancer is one of the most frequently-occurring malignancies in our country. This study is to detect the expression of MAL mRNA, ER, PR and C-erbB-2 (Epidermal growth factor receptor-2) in breast cancer and to analyse the relevance of them, meanwhile to identify the relationship between MAL gene and breast cancer,and to give the test data support for MAL gene becoming an early diagnostic marker which could be used for early clinical diagnosis and evaluation of prognosis.MethodsThe expression of ER (estrogen receptor), PR (progesterone receptor), C-erbB-2 (Epidermal growth factor receptor-2) in breast cancer is detected by SP immunohistochemical assay; MAL mRNA is detected by RT-PCR. The software of SPSS13.0 was used for statistical analysis. Theχ2 test was used for the analysis of the expression of MAL mRNA, ER,PR and C-erbB-2, and Spearman's correlation analysis was used to analyse the relevance among them.Results1. The positive expression rate of ER in breast cancer is 68%, yet that of PR, C-erbB-2 is 56%,52% respectively;2. The positive expression rate of ER in breast cancer trends to decrease accompanying with augment of the tumor. The positive expression rate of ER was 92.86%, 58.62% and 57.14% when the diameter of tumor was ranged within <2cm, 2~5cm, >5cm respectively. The difference among all groups showed great statistical significance(P=0.037). The positive expression rate of ER was 64.10% and 81.82% when the number of lymph node metastasis was≤3 and > 4. There was no statistical significancebetween them(P=0.455). The positive expression rate of ER was 73.08% and 62.50% when the age of patients was 33~50y and 51~83y. There was no statistical significance between them(P=0.423).3. The positive expression rate of PR in breast cancer trends to decrease accompanying with augment of the tumor. The positive expression rate of it was 92.86%,48.28% and 14.29% when the diameter of tumor was ranged within <2cm, 2~5cm,>5cm respectively. The difference was great statistical significanceamong all groups(P=0.001). The positive rate expression of it was 56.41% and 54.55% when the number of lymph node metastasis was≤3 and > 4. There was no statistical significancebetween them(P=0.912). The positive expression rate of it was 76.92% and 33.33% when the age of patients was 33~50y and 51~83y. There showed statistical significance between them(P=0.002).4. The positive expression rate of C-erbB-2 in breast cancer trends to increase accompanying with augment of the tumor. The positive expression rate of it was 14.29%,62.00% and 85.70% when the diameter of tumor was ranged within <2cm, 2~5cm, >5cm respectively. There was great statistical significanceamong all groups(P=0.002). The positive expression rate of it was 53.85% and 45.45% when the number of lymph node metastasis was≤3 and >4. There was no statistical significancebetween them(P=0.623). The positive expression rate of it was 46.15% and 58.33% when the age of patients was 33~50y and 51~83y. There was no statistical significancebetween them(P=0.389).5. The positive expression rate of ER and PR in breast cancer had positive correlation(rs=0.603,P<0.001);The positive expression rate of ER and C-erbB-2 in breast cancer showed negative correlation (rs=-0.451,P=0.001); The positive expression rate of PR and C-erbB-2 in breast cancer showed negative correlation(rs=-0.616,P<0.001).6. The positive expression rate of MAL mRNA in normal breast tissues was 100%, and 20% was in breast cancer. There was statistical significancebetween them(P=0.001). The positive expression rate of MAL mRNA in breast cancer without lymph node metastasis was 0.81±0.21, it was 0.40±0.16 in those patients with lymph node metastasis. There was statistical significancebetween them(P<0.001). The positive expression of MAL mRNA in breast cancer detecting by means of RT-PCR was 0.94±0.21 and 0.51±0.12 when the diameter of tumor was <2cm and≥2cm. There was statistical significancebetween them(P<0.001).Conclusions1. ER,PR and C-erbB-2 all have higher positive expression rate in breast cancer(>50%). All of them could be a good marker for early diagnosis of breast cancer.2. ER has higher positive expression rate in breast cancer. The positive expression of ER in breast cancer has negative correlation with diameter of tumor,but has no correlation with the number of lymph node metastasis and the age of patients.3. PR has higher positive expression rate in breast cancer. The positive expression of PR in breast cancer has negative correlation with diameter of tumor and the age of patients, but has no correlation with the number of lymph node metastasis .4. C-erbB-2 has higher positive expression rate in breast cancer. The positive expression of PR in breast cancer has positive correlation with diameter of tumor but has no correlation with lymph node metastasis and age of patients.5. The positive expression of ER and PR in breast cancer have positive correlation, but they have negative correlation with that of C-erbB-2.6. MAL mRNA has lower positive expression rate in breast cancer compared with that in normal breast tissue. The rate of it has negative correlation with diameter of tumor and lymph node metastasis,but has no correlation with the age of those patients. The positive expression of MAL mRNA and the positive expression of ER or PR in breast cancer have no positive correlation,but that has negative correlation between MAL mRNA and C-erbB-2.
