And to figure out the correlation between c-erbB-2,c-met,nm-23 oncoprotein expression and mRNA expression in gastric cancer tissue. Finally to evaluate the clinical significance of c-erbB-2,c-met,nm-23 genes in tumor diagnosis and state of tumor progression appraisal.Methods:1) 52 cases of gastric carcinoma tissue sample and 42 cases of nontumorous tissue were collected in SHANTOU university medical college, the first affiiated hospital surgery departments and gastrosopy room. After proper disposal, we detected the samples with certain kinds of method and analysis the data.2) C-erbB-2,c-met,nm-23 protein expression in gastric tumor tissue and nontumorous tissue were evaluated cby IHC method.3) We freezed and saved the fresh gasric tumor tissue, and extracted the gross RNA from tissue by TRIZOL, investigated c-erbB-2,c-met,nm-23 gene expression by semi-RT-PCR. The c-erbB-2,c-met,nm-23 gene amplification product was sent to sequencing and analysis.4) All of statistical analysis was performed with statistical package SPSS version 13.0 software.Results:1) Protein positive rates for c-erbB-2,c-met,nm-23 are 55.77%(29/52), 65.38%(34/52),46.15%(24/52), respectively. Comparing with the positive rate of the proteins in nontumorous tissue, 19.05% (8/42),23.81% (10/42),95.24% (40/42) respectively, the proteins expression difference between tumor tissue and nontumorous tissue is significant(P<0.01)。2) C-erbB-2 protein expression is higher in intestinal type group,lymph node metastasis-positive group and serosa invasion-positive group than that in diffuse type group,lymph node metastasis-negative group,serosa invasion-negative group, and the difference is statistically significant (P<0.05)。3) C-met protein expression is higher in lymph node metastasis-positive group and serosa invasion-positive group than that in lymph node metastasis-negative group,serosa invasion-negative group, and the difference is statistically significant (P<0.05).4) Nm-23 protein expression is statistically higher in lymph node metastasis-negative group than that in lymph node metastasis-positive group(P<0.05).5) C-erbB-2 mRNA expression in intestinal type group,serosa invasion-positive group is significantly higher than that in diffuse type group,serosa invasion-negative group(P<0.05).6) C-met mRNA expression in serosa invasion-positive group is significantly higher than that in serosa invasion-negative group(P<0.05).7) Nm-23 mRNA expression in lymph node metastasis-negative group is significantly higher than that in lymph node metastasis-positive group(P<0.05).8) There are positive correlations of c-erbB-2,c-met between mRNA and protein expression, respectively(P<0.01),(P<0.05).Conclusions:1) High protein expression for c-erbB-2 gene and c-met gene in gastric tumor tissue, and low protein expression for rim-23 gene in gastric tumor tissue indicate a diagnosis value for detect these proteins expression in tissue to distinguish nonmalignant and malignant diseases.2) C-erbB-2 protein trend to express in intestinal type gastric tumor and play a role in tumor lymph node metastasis and serosa invasion. C-erbB-2 mRNA trend to express in intestinal gastric tumor and promote tumor development. C-erbB-2 mRNA expression is positive correlation with protein expression, data suggest c-erbB-2 mRNA expression or protetin expression could be used as a index to evaluate gastric tumor progression extent.3) C-met protein is related with tumor lymph node metastasis and invasion. C-met mRNA expression relate with tumor development. C-met mRNA expression is positive correlation with protein expression. Result suggest c-erbB-2 mRNA expression or protetin expression could be used as a index to evaluate gastric tumor progression extent.4) Nm-23 mRNA and protein expression may suppress gastric tumor lymph node metastasis.
