ObjectiveRecently, the incidence of choledocholith increased year by year, it was recognized that biliary sports dysfunction play an important role in the process of choledocholith disease, while it was recognized that the gastrointestinal peptide hormones has certain effect to biliary and contraction and relaxation of Oddi,s sphincter. Gastrointestinal peptide hormones may affect the formation of gallstones by changing one or certain factors. Therefore it is necessary to study these factors in order to explore the cause of choledocholith and provide some help to the treatment. In this study, the content of cholecystokinin(CCK),motilin(MTL) and vasoactive intestinal peptide(VIP) of bile of patients ofcholedocholith are combine detected through radioimmunoassay( RIA) to explore their role and clinical significance in the formation of choledocholith.Methods:Collect hospitalized patients for ERCP treatment due from February 2009 to December 2010, divided these patients into two groups: one group are patients withcholedocholith and the control group are patients with normal ERCP imaging. Collection of specimens:perfect relational examination ,more than 8 hours of fasting followed by the ERCP surgery. Insert the side-view duodenum mirror from mouth into the duodenum papilla, insert the guide wire from the biopsy holes, insert catheter along the guide wire, pump 5ml of bile with sterile syringe, place it in plastic test tube with 30ul 7.5% EDTA and 40ul Aprotinin and shake, centrifuge under 40C (1500 rev / min) for 15 minutes, take the supernatant, and put it in refrigerator of -700C under test. Use radioimmunoassay method:detect the content of cholecystokinin(CCK), motilin(MTL), vasoactive intestinal peptide(VIP) in the samples that collected. Denote the data results with x±S, use SPSS13.0 statistical software for data analysis, do independent sample t test with the data obtained, P <0.05, indicate that the difference is significant.Results 1. Test results of Cholecystokinin(CCK):The test of Cholecystokinin is failed, We can't get a good result. The experimental data of Cholecystokinin(CCK) can't be found in both calculus of common bile duct group and contrast group.2. Test results of Motilin(MTL):The content of Motilin is 690.284±453.607pg/ml in the bile of the patients of calculus of common bile duct. The ordinary content of Motilin is 316.605±171.375pg/ml. The content of Motilin of the patient is higher than the ordinary one, the comparison between the patient and ordinary one is very significant in the sense of statistics (P<0.05).3. Test results of Vasoactive Intestinal Peptide(VIP)The content of VIP is 1807.490±652.103pg/ml in the bile of the patients of calculus of common bile duct. The ordinary content of VIP is 668.876±288.322pg/ml. The content of VIP of the patient is much higher than the ordinary one,the comparison of VIP is also very important in the sense of statistics (P<0.05).Conclusions1. The content of motilin was significantly higher in the bile of the patients of calculus of common bile duct, The results show that the motilin can affect the movement of biliary and result in calculus of common bile duct in some sense.2. The content of vasoactive intestinal peptide was also significantly higher in the bile of the patients of calculus of common bile duct, We also found that vasoactive intestinal peptide can also affect the movement of biliary and result in calculus of common bile duct in some sense through the results.3. The test of cholecystokinin is failed, we can't get useful results.
