ObjectiveThe method of flow cytometry in patient with premature coronary heart in peripheral blood mononuclear cells the expression of haptoglobin ,quantitative analysis of serum haptoglobin expression and early onset coronary heart disease.The study investigated the hapatoglobin in the early-onset coronary heart disease value of the protein with coronary heart disease signs of myocardial injury.Methods1,SubjectsCHD group:60 premature coronary heart disease(32male, aged<55 years; 28 female,aged<65years; mean aged 51.2 years)were consecutively collected from the hos- pitalized in the department of Tai Shan medical college from October 2009~October 2010. All patients were confirmed by coronary angiography of WHO,The ECG, echocardio- graphy,biochemical and other tests ruled out infection,other than thrombotic coronary artery disease,cancer,automimmue diease.Control group:40 control group subjects(23 males and 17 female, aged from 45 to 60 years,mean aged 50.6 years old).All subjects were consecutively collected in the affiliated hospital of Tai Shan medical college.All patients were confirmed normal by coronary angiography. excluded heart disease,cerebral vascular accident,liver and kidney dysfunc–tion coagulation abnormalities and infectious disease by routine ECG, echocardiography and biochemical tests.2,Study methods:2.1 Risk for coronary heart disease: smoking, family ischemic cardiovascular dis- ease,weight,sex, hypertension, type 2 diabetes mellitus.2.2 Blood sampling:Peripheral venous blood samples were collected in the morning after an overnight fast, 2.0ml of heparin anticoagulant set,the other pumping 5.0ml tube set gel clotting, coagulation blood sample was centrifuged (4000r/min for 8 minters), the supernatant was collected and it was conserved at -20℃,the content of haptoglobin in peripheral blood were detected by flow cytometry producted by BD company of USA,Reagents and standard materials were purchased from Beijing Biosynthesis Biotechnology Co.Ltd.The concentration of serum haptogolin were detected by Enzyme -linked immunosorbent assay and the levels of serum hs-CRP were detected by immune turbidimetry by Hitachi 7600-020 automatic biochemical analyzer,the kit was purchased from Hitachi.The contents of sersum D-dimer were detected by immunotur -bidimetric and the kit was purchased from Sysmex Medical Electronics Co,Ltd.3. Statistical analysesStatistical analysis were performed with the SPSS for Windows 18.0 statistical program package.All continuous variables were expressed as mean value±standard deviation (mean±SD), Comparison between groups was performed by t test;χ2 test was used for the comparison among the samples.Non-parametric statistics was used about non-normal distribution and variance of the missing data. P value<0.05 was considered statistically significant.Results:1.1 The premature CHD group patients were more frenquently found in the history of smoking (P=0.005),the family history of ischemic cardiovascular disease(P=0.000)and diabetes(P=0.02),compared with the control group.There were no statistically significant in the hypertention gender,weight.1.2 Comparasion between premature CHD group and control group:The concentration of peripheral blood haptoglobin,serum haptoglobin,hs-CRP and D-dimer in premature coronary heart disease group was significantly higher, than the control group, the difference between the two groups was statistically significant.1.3 The correlation analysis between the concentrations of peripheral blood hapto- globin,serum hs-CRP and D-dimer among the patients with premature coronary heart disease:The analysis showed between premature CHD group and control group: The concentrations of haptoglobin was positive correlated with the concentration of hs-CRP (r=0.841,P<0.01). The concentrations of haptoglobin was positive correlated with the concentration of D-dimer(r=0.795,P<0.01)1.4 The correlation analysis between serum haptoglobin,hs-CRP and D-dimer among the patients with premature coronary heart diseaseThe analysis showed between premature CHD group and control group: The concentrations of haptoglobin was positive correlated with the concentration of hs-CRP (r=0.928,P<0.01).The concentrations of haptoglobin was positive correlated with the concentration of D-dimer(r=0.752,P<0.01)Conclusion:1.1 Smoking ,family ischemic cardiovascular disease,diabetes are the risk factors of premature coronary disease.1.2 Haptoglobin,hs-CRP and D-dimer were over expressed in the premature heart disease group compared to healthy group.1.3 Haptoglobin involved in immuneregulation in premature coronary disease. Haptoglobin is the protein markers of myocardial injure of premature coronary disease.1.4 The combined detection of haptoglobin,hs-CRP and D-dimerin will play crucial role in diagnosis, identification,assessment,treatment,prognosis of premature coronary heart disease.The combined detection can prevent the occurrence of cardiovascular disease ,and thus play a greater clinical role.
