Background:Chronic urticaria and chronic eczema are allergic diseases caused by many factors, with characteristics of complex etiology, different length of clinical course, repeated outbreak, and difficult to prevent. TCM holds that, although both of them assume the epidermis suffering, they are closely linked with viscera and blood. Usually they show clinical symptoms such as wheal, skin lesions, vesicle erosion, pruritus, etc. So it makes sense to explore the pharmacodynamics effect and its relevant mechanism of an Chinese herba preparation for oral administration with the main actions of dispelling wind and arresting itching, clearing heat and cooling blood, and xeransis.Qufengzhiyang Granule, as a common proved recipe of a famous doctor, is consist of nearly ten herbs. It is used to treat wind-heat chronic urticaria, cutaneous pruritus, damp-heat type chronic eczema, etc. We have surveyed vast literature and ancient books, summarized, sorting and sublimation those effective methods and formula.Through the pharmacodynamics experiment and related mechanism discussion, we herein study the main pharmacologic action and relevant mechanism of Qufengzhiyang Granule to provide theoretical guidance for clinical practice. Methods:1.Drugs:Qufengzhiyang Granule, provided by the Oriental Medicine Lin Pharmaceutical Co., LTD. Piminxiao capsule, Shenyang Aohua Pharmaceutical Co., LTD. Chlorpheniramine, Guangzhou Kanghe Pharmaceutical Co., LTD. Dexamethasone Acetate, Zhejiang Xianju Pharmaceutical Co., LTD.2. Animals:KM mice, SPF level,18-22g, bisexual each half; SD rats, SPF level,160-200g, bisexual each half; SD rats, SPF level,90-100g, male. Guinea pig, clean grade, 200-300g, bisexual each half. All animals are provided by the Animal Center of Southern Medical University. Qualified SCXK number:guangdong 2006-0015.3.Outcome measures:3.1 The experimental study on allergy resistance of Qufengzhiyang Granule3.1.1 Rats to the same passive influence of skin allergic experiment3.1.2 Surveying the influence of Qufengzhiyang Granule on rat's skull periost mast cells degranulation3.2 Surveying the effects of Qufengzhiyang Granule on urtiearia with non-immunity3.2.1 Effect on dimethyl sulfoxide-induced guinea pig ear swelling3.2.2 Effect on Cinnamic acid-induced guinea pig ear swelling3.3 Surveying the effects of Qufengzhiyang Granule on exogenous drug skin itching3.3.1 Effect on dextran-induced mouse skin itching3.3.2 Effect on histamine-induced guinea pig skin itching3.3.3 Effect on 4-aminopyridine-induced mouse skin itching3.4 To observe effect on anti Inflammation of Qufengzhiyang Granule3.4.1 Effect on capillary permeability in rats. 3.4.2 Effect on xylene-induced mouse ear swelling3.5 To explore the mechanism of anti-allergy3.5.1 To study the change of IFN-yin serum of type I hypersensitivity rat3.5.2 To study the change of IL-4 in serum of type I hypersensitivity rat3.5.3 To study the change of IgE in serum of type I hypersensitivity rat4. Statistical analysis:Software SPSS 13.0 was used for statistical analysis. The experimental data were indicated by mean±tandard deviation(X±s). Repeated measures variance analysis and the single-factor analysis of variance(ANOVA) were used to analyze the data. LSD method was used in multiple comparisons between groups when the variance is homogeneous. Dunnett's T3 method was used when variance is not homogeneous. P<0.05 indicated that the difference have statistical significance.Results:1. The experimental study on allergy resistance of Qufengzhiyang Granule1.1 Rats to the same passive influence of skin allergic experimentThe data of rats skin optical density in antisera injection site were analyzed by one-way ANOVA. It is shown that there was significant difference among groups(F=3.776, P=0.005) in 1:1 antisera injection site, LSD method was used for further analysis; and there was no significant difference among groups(F= 1.922, P=0.106) in 1:2 antisera injection site. In the PCA 1:1 antisera injection site, compared with the model group, the optical density in the high-, medium- and low-dose Qufengzhiyang Granule groups, chlortrimeton group and Piminxiao capsule group were significantly decreased(P<0.05).1.2 Surveying the influence of Qufengzhiyang Granule on rat's skull periost mast cells degranulationThe data of rat's skull periost mast cells degranulation rate were analyzed by one-way ANOVA. It is shown that there was significant difference among groups(F=154.57, P=0.000), LSD method was used for further analysis. In the rat's skull periost mast cells degranulation rate, compared with the model group, the nomal group, the high-, medium- and low-dose Qufengzhiyang Granule groups, chlortrimeton group and Piminxiao capsule group were significantly decreased (P< 0.05).2. Surveying the effects of Qufengzhiyang Granule on urtiearia with non-immunity2.1 Effect on dimethyl sulfoxide-induced guinea pig ear swellingThe data of guinea pig ear swelling degree were analyzed by two-way analysis of variance with repeated meseasures:there was no interaction between the time point and the groups(F=0.762, P=0.716), main effects between groups (F=3.548, P=0.008) and main effects between the time groups (F=121.580, P=0.000). In tne guinea pig ear swelling degree, compared with the model group, without the 45 min,180 min low-dose Qufengzhiyang Granule groups and the 120 min medium- and low-dose Qufengzhiyang Granule groups, the rest of the groups can reduce the ear swelling degrees (P<0.05).2.2 Effect on cinnamic acid-induced guinea pig ear swellingThe data of guinea pig ear swelling degree were analyzed by two-way analysis of variance with repeated meseasures:there was interaction between the time point and the groups(F=3.746, P=0.000), main effects between groups (F=8.773, P=0.000) and main effects between the time groups (F=497.646, P=0.000). In tne guinea pig ear swelling degree, compared with the model group, without the 15 min,30 min groups, the 120 min low-dose Qufengzhiyang Granule groups and the 180 min medium-and low-dose Qufengzhiyang Granule groups, the rest of the groups can reduce the ear swelling degrees (P<0.05)3. Surveying the effects of Qufengzhiyang Granule on exogenous drug skin itching 3.1 Effect on dextran -induced mouse skin itchingThe data of mouse skin itching frequency and duration within 30min were analyzed by one-way ANOVA. It is shown that there was significant difference among groups on mouse skin itching frequency (F=5.662, P=0.000) and mouse skin itching duration(F=2.603, P=0.024). LSD method was used for further analyze. In the dextran-induced mouse skin itching, compared with the model group, the nomal group, the high-, medium- and low-dose Qufengzhiyang Granule groups, chlortrimeton group and Piminxiao capsule group were significantly decreased(P< 0.05), could decrease scratching frequency and duration.3.2 effect on histamine-induced guinea pig skin itchingThe data of the times and dosage of daubing histamine on guinea pig were analyzed by one-way ANOVA. It showed that there was significant difference among groups on the times (F=8.073, P=0.000) and dosage(F/welch=12.935, P=0.000). LSD method was used for further analyze. In the histamine -induced guinea pig skin itching, compared with the model group, the dosage and times of histamine daubing on the high-, medium- and low-dose Qufengzhiyang Granule groups, and chlortrimeton group were significantly increased(P<0.05), without Piminxiao capsule group.3.3 Effect on 4-aminopyridine -induced mouse skin itchingThe data of 4-aminopyridine-induced mouse skin itching frequency and duration within 30min were analyzed by one-way ANOVA. It showed that there was significant difference among groups on mouse skin itching frequency (F=3.905, P=0.004) and mouse skin itching duration(F=9.631, P=0.000). LSD method was used for further analysis. In the 4-aminopyridine -induced mouse skin itching, compared with the model group, frequency of the high-, medium- and low-dose Qufengzhiyang Granule groups, chlortrimeton group and Piminxiao capsule group were significantly decreased(P<0.001), while without low-dose Qufengzhiyang Granule, duration of the rest group were all significantly increased(P<0.01).4. To observe effect on anti-inflammation of Qufengzhiyang Granule4.1 Effect on capillary permeability in ratsThe data of rat's skin optical density were analyzed by one-way ANOVA. It showed that there was significant difference among groups(F=11.390, P=0.000), LSD method was used for further analysis. In the rat's capillary permeability, compared with the model group, optical density of the high-, medium-and low-dose Qufengzhiyang Granule groups, chlortrimeton group and Piminxiao capsule group were significantly decreased(P<0.01).4.2 Effect on xylene-induced mouse ear swelling degreeThe data of mouse ear swelling degree were analyzed by one-way ANOVA. It is shown that there was significant difference among groups(F=4.873, P=0.000), LSD method was used for further analysis. In the mouse ear swelling degree, compared with the model group, the ear swelling degree of the high-dose Qufengzhiyang Granule groups, chlortrimeton group and Piminxiao capsule group were significantly decreased(P<0.01).5. To explore the mechanism of anti-allergy5.1 To study the change of IFN-yand IL-4 in serum of typeⅠhypersensitivity ratThe data of IFN-ycontent in serum of typeⅠhypersensitivity rat were analyzed by one-way ANOVA. It is shown that there was significant difference among groups(F=3.111, P=0.010), LSD method was used for further analysis. In the IFN-y content in serum of type I hypersensitivity rat, compared with the model group, the normal group, high-, medium-, low-dose Qufengzhiyang Granule groups, chlortrimeton group and Piminxiao capsule group were significantly different(P< 0.05).The data of IL-4 content in serum of type I hypersensitivity rat were analyzed by one-way ANOVA. It is shown that there was significant difference among groups(F=3.468, P=0.005), LSD method was used for further analysis. In the IL-4 content in serum of typeⅠhypersensitivity rat, compared with the model group, the normal group, high-, medium-dose Qufengzhiyang Granule groups, chlortrimeton group and Piminxiao capsule group were significantly different(P<0.05).The data of IFN-γ/IL-4 content in serum of typeⅠhypersensitivity rat were analyzed by one-way ANOVA. It showed that there was significant difference among groups(F=6.118, P=0.000), LSD method was used for further analysis. Compared with the model group, the normal group, high-, medium-dose Qufengzhiyang Granule groups, chlortrimeton group and Piminxiao capsule group were significantly different(P<0.01).5.2 To study the change of IgE in serum of typeⅠhypersensitivity ratThe data of IgE content in serum of typeⅠhypersensitivity rat were analyzed by one-way ANOVA. It is shown that there was significant difference among groups(F=11.343, P=0.000), LSD method was used for further analysis. In the IgE content in serum of type I hypersensitivity rat, compared with the model group, the normal group, high-, medium-,low-dose Qufengzhiyang Granule groups, chlortrimeton group and Piminxiao capsule group were significantly different(P< 0.01).Conclusions:1. Qufengzhiyang Granule has antiallergic effects:it can antagonize guinea pig urtiearia with non-immunity, inhibit rats passive cutaneous anaphylaxis, restrain rat's skull periost mast cells degranulation.2. Qufengzhiyang Granule has anti-pruritic effect:it can inhibit dextran and 4-aminopyridine-induced mouse skin itching, increase the tolerance histamine phosphate on guinea pig.3. Qufengzhiyang Granule has anti-inflammatory properties:it can reduce xylene-induced mouse ear swelling and histamine-induced capillary permeability increasing.4. Qufengzhiyang Granule can adjust corresponding cytokines of the typeⅠallergy, such as IL-4,IFN-γIgE; and has effect of anti-typeⅠallergy.
