Objective To establish the PC12 cell LPS damage model and the primary neurons LPS damage model, and use this two kinds of LPS damage model to discuss GM1 and NGF whether have the function to repair the damage, and to find its mechanism. It may have same relations with nuclear transcription factors (NF-KB) activation. Method (1) the establishment of LPS damage model: culture the PC12 cell with the different concentrations of LPS injury, and then determine by MTT method to make sure the damage concentration; Take the cortex of new born SD mice, separating out the original generation neurons to culture in the different concentrations of LPS injury, and then determine by MTT method to make sure the damage concentration. (2) The protect experiment:after determined LPS injury, we culture the cells in the different concentrations of NGF, GM1. And then we assay the cells survival by MTT and observe the cellular morphology to determine the best concentration. At last, we assay the content of NF-KB by RT-PCR. Meanwhile, join in D-PDMP to inhibitor the endogeneous GM1, and then use the same way. Try to clear the protective effects and mechanism of LPS injury by GM1 and NGF, and wether have some relations with NF-KB. Result (1) through these experiments, we find that the LPS injury has certain concentration dependence, and select the concentration of 100nmol/L. (2) through the morphological observation, MTT and RT-PCR analysis, we find that GM1 and NGF can protect the LPS injury. And use D-PDMP to inhibitior the endogeneous GM1, we can find the same effect. Conclusion GM1 and NGF have proective to LPS injury, and they also can repair the injury. The mechanism may have some connect with the activation of NF-KB signaling pathways.
