Objective: To investigate the expression of AKT and PTEN protein and their relationship with clinic characteristic of esophageal carcinoma.Methods: The expression of AKT and PTEN protein in 80 patients with esophageal cancer,12 case of high-level intraepithelial neoplasia ,20 case of low-grade intraepithelial neoplasia,12 case of normal esophageal mucosa were examined by immunohistochemical S-P technique. At the same time , relationship between AKT and PTEN in esophageal cancer was explored.Results:1. The expression of AKT and PTEN Protein in different esophageal tissues. The Positive rate of their expression in 80 cases of esophageal cancers,12 case of high-level intraepithelial neoplasia,20 case of low-grade intraepithelial neoplasia and 12 case of normal esophageal mucosa tissues were 55.0%(44/80),25.0%(3/12),10.0%(2/20),8.3%(l/12) and 45.0%(36/80),66.7%(8/12),80.0%(16/20),100%(12/12).There were significant differences among the expression of AKT and PTEN in different esophageal tissues (P=0.000).The expression of AKT in esophageal cancer group was higher than in benign and normal groups (P<0.001).The expression of PTEN Protein in esophageal cancer group was lower than in benign and normal group (P<0.001).2. The relationship between their expression (AKT and PTEN Protein) and clinicopathologieal factors. The positive rate of AKT in well, moderate differentiation cancer group (G1+G2) was 44.2%(23/52).It is lower than that in Poor differentiation cancer group (G3).The positive rate was75.0%(21/28).There was significant difference between these two groups (P=0.008).The positive rate of PTEN protein expression in these groups were respectively 55.8%(29/52) and 25.0%(7/28) .There were significant difference between them(P=0.008).The positive rate of AKT protein expression in stageⅠ/Ⅱof EC were respectively 32.6%(15/45) and 80.6%(29/35).There were significant difference between them (P<0.01).The positive rate of PTEN protein expression in these groups were respectively 53.3%(25/45),28.5%(11/35).There were significant difference between them (P<0.01). The positive rate of AKT protein expression in positive or negative lymph node metastasis of EC were respectively 71.8% (28/39) ,39.0%(16/41).There were significant difference between them(P<0.01).The positive rate of PTEN protein expression in these groups were respectively 28.2%(11/39),60.1%(25/41).There were significant difference between them (P<0.01).Both of AKT and PTEN were not related with age,sex and size of tumor.(P>0.05)3. The correlation between the expression of AKT and PTEN protein in EC. Their expression which were all positive or negative were respectively 11 cases. The expression of AKT was Positive with negative expression of PTEN were 33 cases. The expression of PTEN was Positive with negative expression of AKT were 25 cases. There was inversely correlation between the expression of AKT and PTEN Protein in EC (r=-0.444 P<0.01)Conclusion:1. The activation of AKT protein and the absence of PTEN were correlated with carcinogenesis,progression,invasion and metastasis of EC.2.
