Objective: To study the therapeutic effect and its possible mechanisms of Tiaozhi granules on hyperlipidemic fatty liver in rats.Methods: Rat model with hyperlipidemic fatty liver was established by feeding the rats with fatty milk. Then effects of tiaozhi granules on lipid level, liver function and antioxidative index in hyperlipidemic fatty liver were observed respectively. Histopathological changes of rat liver and ultrastructure were observed under light and electron microscopy. The expressions of peroxisome proliferator-activated receptorα/γ(PPARα/γ), cholesterol 7α-hydroxylase (CYP7A1), scavenger receptor B1(SR-B1) mRNA and protein, liver fatty acid-binding protein(L-FABP ), diacylglycerol acyltransferase (DGAT), sterol regulatory element binding protein 1c/2 (SREBP-1c/2) and low density lipoprotein receptor (LDLR) mRNA were inspected by RT-PCR and Western-blot methods, respectively.Results: After treatment with tiaozhi granules 0.3-1.2 g/kg for 6 weeks, the coefficient of hepatic weight, serum TC,TG,FFA,LDL-C,AST,ALT,MDA and hepatic TG, TC, FFA and MDA levels were significantly decreased(P<0.05 or P<0.01), while the serum levels of HDL-C and GSH and the hepatic levels of SOD and GSH were obviously increased(P<0.01)with a certain trend of dose-dependence, and under light and electron microscopy the degree of hepatocyte steatosis was lessened to a large extent (P<0.01). According to the results of RT-PCR, tiaozhi granules could significantly increase the PPARα/γ, L-FABP, CYP7A1 and SR-B1mRNA expressions(P<0.05 or P<0.01), and decrease the SREBP-1c/2,DGAT and LDLR mRNA expressions in hepatic tissue in hyperlipidemic fatty liver rats(P<0.01). Tiaozhi granules could also increase the hepatic PPARα/γ, CYP7A1 and SR-B1 protein expressions in hyperlipidemic fatty liver rats(P<0.01).Conclusion: Tiaozhi granules have certain therapeutic effect on hyperlipidemic fatty liver in rats, and the mechanism might be related to its ability to regulate the CYP7A1, SR-B1, L-FABP, DGAT, and LDLR expressions via activating the PPARα/γand subsequently inhibiting SREBP-1c/2 expression, and to enhance antioxidation.
