Objective: To investigate the mechanisms of Osteopontin and CD44 in stone formation .Methods: 1.Hyperoxaluria rat model was established by administration of 0.5% ethylene glycol and 0.5% ammonium chloride in drinking water for 3 to 28 day. 2.Calculus crystal deposited in renal tubular were observed under light microscopy (HE-staining). 3.Calcium mineralization in the basement membrane of renal tubular epithelial cells and interstitium were Observed by calcium-staining in kidney samples. 4.The expression and location of osteopontin and CD44 in kidney were observed by immunohistochemistry. 5.The relationship among the expression of osteopontin, CD44 and calculus crystal on different time were analyzed. investigate the role of Osteopontin and CD44 in stone formation.Results: The expression of osteopontin and CD44 increase in accordance with the time and the deposition of Calculus crystal,later attenuated.Conclusions:Under the hyperoxaluria stimulating,the expression of the OPN and CD44 increase in accordance with the deposition of calculus crystal. the OPN and CD44 keep the positive correlation,which maybe the source of Randall Plaque.
