Objective:To investigate the mechanisms of calculus crystal transport by macrophage in kidney.Methods:1.Hyperoxaluria rat model was established by administration of 1%ethylene glycol and 1%ammonium chloride in drinking water for 3 to 14 days.2.24h rat urines were collected in metabolism cage,urinary oxalate were analyzed by ion chromatography.Urinary crystals were observed under light microscopy.3.Calculus crystal deposited in renal tubular were observed under light microscopy(HE-staining).4.Calcium mineralization in the basement membrane of renal tubular epithelial cells and interstitium were observed by calcium-staining in kidney samples.5.The expression and location of osteopontin and macrophage in kidney were observed by immunohistochemistry.6.Macrophage and calculus crystal at the basement membrane of renal tubular epithelial cells and interstitium were observed under the transmission electron microscope.The relationship among the expression of osteopontin,macrophage and calculus crystal on different time were analyzed.Results:1.The expression of osteopontin increase in accordance with the excretion of oxalate in urine(r=0.887,P<0.05),osteopontin expression is from the collecting tube to loops of Henle.2.The macrophage in the basement membrane of renal tubular epithelial cells and interstitium increase in accordance with the expression of osteopontin(r=0.596,P<0.05).3. Macrophage located in the basement membrane of loops of Henle expressed early than that of calculus crystal,and the deposition of calculus crystal increase in accordance with the macroghage.4.The macroghage moved from vessel to the basement membrane of loops of Henle in kidney,then disrupted and released the calculus crystal.Conclusion:Our datas support that macrophage take part in the calculus crystal transport in kidney at the basement membrane of loops of Henle,which may be the source of Randall plaque.This process may be mediated by osteopontin.
