| Object: To detect the methylation status of P16 gene of the Uyghur,s cervix squamous cell carcinoma and normal cervical issues ,and explore the correlation between P16 gene CPG methylation sites and the cervix squamous cell carcinoma in Uyghur women. Methods:(1)Using MALDI-TOF MS to detect P16 gene promotor CPG methylation sites of 20 specimens of cervix squamous cell carcinoma and 20 specimens of normal cervical issues in Uyghur women.(2) The infection state of HPV 16 was detected by using Polymerase chain reaction(PCR) in different two groups. Results: (1) Among the 16 CPG sites in P16 gene promoter area, CPG1-2, CPG 6 sites were different in two groups ,the levels of CPG1-2, CPG6 methylation sites in cervix squamous cell carcinoma were higher than those in the normal group; (2) The infection ratio of HPV16 in cervix squamous cell carcinoma was higher than that in the normal group with sifnificant difference statistically (P<0. 05) ; (3) P16 gene methylation among these sites in cervix squamous cell carcinoma group was not associated with HPV16 infection. Conclusion:P16 gene CPG 1-2, CPG 6 metnylation may be related to the occurrence of the cervix squamous cell carcinoma in Uyghur women. but not associated with HPV16 infection;HPV 16 infection and the cervix squamous cell carcinoma in Uyghur Women was correlated. |
