Objective:To investigate the influence of pulp tissue-derived growth factor TGF-β1 expression by external stimulation and the formation of tertiary dentin, discussion the correlation between endogenous growth factor TGF-β1 and the formation of dentin pulp complex in situ.Method:6 dogs,12-24months old, were selected for experiment, four molars were chosen for each dog, and were divided into A,B,C and D group randomly. A considered as the control, and B,C,D were experimental groups, which were prepared 2,2.5 and 3mm depth, respectively, but the width and height were all 3mm, all above were considered as the external stimulation. All the cavities were temporary closed by zinc oxide eugenol. The teeth were removed for 2,4,and 6 weeks, tissue section were prepared for observation the formation of the third phase of dentin by HE staining and the expression of TGF-01 by Immunohistochemistry.Results:The third phase of dentin was not observed with 2.5mm depth stimulation after 2 weeks, however, the tissue sections were showed the vacuolar degeneration of odontoblasts and the reparative dentin and predentin were formed with 3 mm depth stimulation after 4,6 weeks. The TGF-β1 of A,B,C,D group were expressed, and the intensity was increasing with the time and stimulation increased, there were significant differences(P<0.05).Conclusion:The expression of endogenous growth factor TGF-β1 was increased by external stimulation, furthermore, TGF-β1 involved in the formation of dentin pulp complex, this information may be a desirable option to summarize that the stimulus intensity and time were positive correlation to the expression of endogenous growth factor TGF-β1 and the formation of dentin pulp complex.
