Influences Of Dual Blockage Of Rennin Angiotensin System On Renal Expression Of Vascular Endothelial Growth Factor And Endostatin In Type 2 Diabetic Rats

Objective: To investigate the effect of combined use of ACEI and ARB on renal expression of vascular endothelial growth factor and endo- statin in type 2 diabetic rats,and their possible mechanisms。Methods : Sixty SD rats were randomly divided type 2 diabetic group ( n = 52) and control group (n =8), A was normal group toreceive normal feeding + buffer solution ,otherts receive highlipids and high- sucrose feeding + streptozotocin ( STZ) group.After model established, the fifty-two SD rats were randomly divided into four groups:Diabetic nephropathy model group(group B),treat with irbesartan (group C),treat with benazepril (group D),treat with irbesartan + benazepril (group E)。After 8 weeks, urea nitrogen,serum creatinine,24 hour total Protei nurine,blood glocoses,kindney weight/body weight were measured。Expressions of vascular endothelial growth factor and endostatin were detected by immuno-histochemical staining。Results: Urea nitrogen,serum creatinine,24 huor total Prote- inurine,blood glocoses,kindney weight/body weight,expression of VEGF,expression of ENS in diabetic rats and treated groups were signif- yciently higher than the normal control group。Urea nitrogen,serum createnine,24 huor total Proteinurine,kindney weight/body weight,expression of VEGF in treated groups were significiently lower than nontreat rats.There were great depression in Urea nitrogen,serum creatinine,24 hour total Proteinurine,kindney weight/body weight,expression of VEGF between E group and C group or D group .but expression of ENS in treated groups were significiently Higher than nontreat rats。It was significiently higher in E group than C group or D group.Conclusions: The imbalance of the expressions between VEGF and ENS in diabetic rats.It could delay the occurrence and development of DN Pathological process.Irbesartan and Benazepril has renal protective effect partly through reducing VEGF expression and improving ENS expression .Combination of both offer futher benefits arrest proteinuria and protect from renal function and structure impairment.
