PurposeTo test Insulin receptorα,βin various types of lung cancer and its clinical relations, so as to lung cancer provide a broader approach.MethodsThe immunohistochemical method in 60 cases of lung cancer, including surgery; small cell lung cancer (SCLC), non-small cell lung cancer (NSCLC, including squamous cell carcinoma, adenocarcinoma, large cell carcinoma) and 40 of its adjacent and 10 cases of normal lung tissue of insulin receptorα(Insulin receptorα) and insulin receptor (insulin receptorβ) detection and analysis of its expression with age, sex, cell type, tumor grade, clinical stage, lymph node metastasis .ResultsInsulin receptorα,β(ISRα,β) distributed in the cell membrane, showed diffuse distribution of positive expression of brown particles. Lung Cancer ISRα,βpositive rates were 75.54%, 70.77%, respectively, higher than the adjacent tissues of 26.54%, 25.09% (p <0.05) and normal lung tissue of 10.78%, 9.9%. (P <0.05) . In adenocarcinoma, squamous cell carcinoma, small cell carcinoma, lung cancer tissues and normal tissues were expressed. ISRα,βhighest expression in adenocarcinoma (p <0.05), in squamous cell carcinoma, small cell lung cancer and adjacent tissues weaker, in the normal lung tissue is the weakest. ISRα,βexpression with sex, age, degree of differentiation (p> 0.05); NTM stage (Ⅲ+ IV) of lung tissue ISRα,βprotein were higher than (I +Ⅱ) period (p <0.05) and lung cancer with lymph node metastasis was significantly higher than those without lymph node metastasis (p <0.05). ISRα,βin lung tissue protein levels were positively correlated (Rs' = 0.623, p <0.05).Conclusion(1) lung cancer tissues, normal lung tissues ubiquitous cell membrane insulinα,βreceptor (ISRα,β), insulin-PI3K / Akt pathway and the Ras / ERK pathway to provide carrier. (2) insulin and insulin receptor binding conformational changes caused by a series of chemical reactions which led to insulin responsiveness and the expression of cell surface receptors has some related sexual. Insulin-α,βreceptors in cells may cause abnormal expression of insulin signaling pathway, especially the significant changes in downstream signaling, thus affecting the metabolism of insulin, promoting cell proliferation and apoptosis regulation。(3) insulinα,βin lung tissue expression of the highest rates of 75.54%, 70.77%, respectively, 26.54% higher than the adjacent tissues, 25.09% and 10.78% of normal lung tissue, 9.9%, suggesting that it may be the factors promoting the development of lung cancer。(4) insulinα,βin expression with age, sex, degree of differentiation, and cell type, clinical stage, lymph node metastasis, prognosis of lung cancer can provide some reference。(5) role in guiding clinical treatment, some of the application of targeted drugs that we provide a reference, consider the application of certain inhibitors, specific blocking or interfering with insulin signaling pathways in cells for the treatment of lung cancer to provide new ideas.
