Objective To investigate the migration effects of human umbilical vascular endothelial cells(HUVECs)in vitro interfered by Endostar in different concentrations and the expressions of miR-126,miR-221,miR-222 of HUVECs, and to explore the anti-angiogenesis mechanism of Endostar by microRNAs.Methods The effect on the migration of HUVECs induced by vascular endothelial growth factor (VEGF), which were interfered with different concentrations of Endostar(50,100,200,500ng/ml) , were determined by cell scratch healing assay. The expressions of miR-126,miR-221and miR-222, which were interfered with different concentrations of Endostar(25,50,100,200,500ng/ml) on different time(1/2,1,2,4,8h), were determined by real-time fluorescent quantitation RT-PCR. To identify the effect on microRNAs in HUVECs modulated by Endostar, relative gene expression data were analyzed by 2-△△CT method.Results①Endostar of different concentrations can inhibit the migration of HUVECs directly and reduce effect of up-regulated migration of HUVECs induced by VEGF. It was in dose dependent manner.②Over expressions of miR-126,miR-221 and miR-222 were detected in the HUVECs.③The expression of miR-126 was down-regulated by Endostar significantly(0.07±0.01~0.56±0.04,P﹤0.01), as compared with that interfered by Endostar before. While the expression of miR-221 and miR-222 were obviously up-regulated by Endostar(1.45±0.06~5.12±0.48,1.42±0.19~6.68±1.51,P﹤0.01), as compared with that interfered by Endostar before.④To the extent of obvervation, the expressions of miR-126,miR-221 and miR-222 was not in dose/time dependent manner by Endostar regulating.Conclusion Endostar can inhibit the migration of HUVECs and can regulate the expressions of miR-126,miR-221 and miR-222 in the HUVECs. Endostar may carry out anti-angiogenesis by its modulating microRNAs.
