ObjectiveTo evaluate the anti-angiogenic effect of endostar for rabbit portal veinVX2tumor thrombus through the correlation study between CT perfusion datameasured before and after treatment and VEGF expression measured byImmunohistochemistry.Methods1.After the recovery of VX2tumor,VX2tumor were inoculated in1rabbitinguinal area,14days later, MRI scan confirmed tumor passage successfully.Remove the passage rabbit inguinal muscle tumor, cut into tumor bar, tumorwere implanted in the18rabbits in the portal vein through open operationmethod,2weeks later, GE Light speed16spiral CT enhanced scan confirmedtumor implantation successfully.18tumor-bearing rabbits were randomlydivided into endostar group,saline group,blank group, each group underwentGE Light speed16spiral CT perfusion scans before and after2weekstreatment, the CT perfusion images and perfusion parameters: blood flow (BF),blood volume (BV), capillary permeability surface (PS) were obtained beforeand after the treatment.2.Blank group rabbits sacrificed after devide group, Endostar group andsaline group rabbits after2weeks of treatment, then sacrificed after CTperfusion scan, and try to keep the same slice with perfusion slice, all thetransplanted tumor tissue fixed by10%Formalin, embedded in paraffin,5μmcontinuous slices, on tumor areas: HE staining, immunohistochemical staining. Immunohistochemistry using SP two step method, VEGF was determinedaccording to the method of literature. Finally, CT perfusion parameters and theimmunohistochemical expression of VEGF results were statistically analyzed.ResultsEach group were obtained satisfied MSCT perfusion imaging, perfusionparameters and expression of VEGF image.1.After treatment, endostar group portal vein tumor thrombus MSCTperfusion parameters:BF, BV, PS were significantly decreased(p<0.05).Endostar group after treatment compared with the saline group of portal veintumor thrombus treatment all CT perfusion parameters: BF, BV, PS weresignificantly decreased (p<0.05).2.Before and after treatment of Endostar,the Immunohistochemistrydetection of portal vein tumor thrombus in the expression of VEGF wassignificantly decreased, the difference was statistically significant (p<0.01).3.The blank group, Endostar group after therapy, saline group aftertherapy of portal vein tumor thrombus CT perfusion parameters of BV wassignificantly correlated with PS, the correlation coefficient r are respectively0.895,0.903,0.873(p<0.01).4.The blank group, portal vein tumor thrombus perfusion parameters andexpression of the VEGF showed highly positive correlation, r values were0.853,0.896,0.813(p<0.01),saline group therapy of portal vein tumorthrombus of the perfusion parameters and expression of VEGF showed highlypositive correlation, r values were0.861,0.892,0.814(p<0.01).Endostargroup therapy of portal vein tumor thrombus of the perfusion parameters andexpression of VEGF showed highly positive correlation, r values were0.863,0.828,0.837(p<0.01).Conclusions1.Endostar as a drug targeting against VEGF,Endostar role in portal veintumor thrombus organization so that the expression level of VEGF of portal vein tumor thrombus in lower.2.MSCT perfusion imaging analyze the perfusion parameters (BF,BV,PS)of portal vein tumor thrombus, can indirectly obtained the the degree ofexpression of VEGF in portal vein tumor thrombus, and then ensure theangiogenesis of tumor clearly, MSCT have a certain value evaluation ofguidance on therapeutic effect of portal vein tumor thrombus.
