ObjectiveMany studies support the concept that a similar state of immune disorders has been described after numerous forms of sepsis. This state is typified by a loss of delayed-type hypersensitivity(DTH) response and an inability to handle secondary infectious challenges, thus predisposing the patient to an increased chance of developing a nosocomial infection,and multiple organ dysfunction syndrome(MODS)is the most frequent complications and the most common cause of death in most of those patients,so if immune disorders could be early intervened or its course was blocked,it would be a great help for improving the prognosis of sepsis. Most of the present concepts are agreed with that the imbalance of T-cell's support-suppress network after sepsis caused by many factors for the immune regulation is the core and foundation of different potential mechanisms of the immune suppression after sepsis. NKT cells,as one subset of the Tregs, except for its cytotoxic activity, can act more importantly as a "brake" (especially in the liver tissue) caused by the local inflammation excessive.This study is to explore the roles of NKT cells in the mechanism of immune disorders after sepsis through studing the liver tissue frequency of NKT cells between a mouse model of sepsis and sham mice.MethodsWe selected 8-10 weeks'C57BL / 6 mice, male. Sepsis model were induced by cecal ligation and puncture (CLP) , while sham mice by anesthesia. In order to observe the function of NKT cells in immune disorders after sepsis, the ratio and apoptosis of NKT cells ,and the radio of the secretion of Th1 / Th2 inflammatory factor(IFN-γ/IL-4)of NKT cells were detected by Flow cytometric analysis after separating the liver tissue of sepsis mice and sham mice,Result1. Compared with the sham mice, the ratio of NKT cells in the liver tissue of sepsis mice decreased progressively in the 24 hours after sepsis.2. Compared with the sham mice, the apoptosis of NKT cells in the liver tissue of sepsis mice increased progressively in the 24 hours after sepsis.3. Compared with the sham mice, the secretion of IFN-γand IL-4 increased, and the radio of the IFN-γ/IL-4 increased obviously, indicating that the functions of NKT cells tend to proinflammatory cytokine responsesConclusion1.
